摘　　要：目的:分析Serpin家族H成员1(SERPINH1)在非小细胞肺癌(NSCLC)中的表达及其作用机制。方法:选取2022年1月-2023年12月于句容市人民医院进行肿瘤切除手术的60例NSCLC患者的肿瘤组织和癌旁组织进行研究。从癌症基因组图谱(TCGA)数据集获得NSCLC组织和正常组织的RNA测序(RNAseq)数据和相应的临床信息,免疫组化评估NSCLC组织中SERPINH1表达,并进行体外细胞实验分析SERPINH1对人肺腺癌细胞系A549细胞增殖、迁移和侵袭的影响。结果:与正常组织相比,SERPINH1 mRNA在NSCLC组织中表达显著上调。SERPINH1高表达患者总生存期和疾病特异性生存期短于SERPINH1低表达患者。NSCLC组织SERPINH1蛋白表达水平高于癌旁组织(P<0.001)。SERPINH1过表达组A549细胞增殖、迁移和侵袭能力高于空白组(P<0.001),SERPINH1敲低组A549细胞体外增殖、迁移和侵袭能力低于空白组(P<0.001)。结论:SERPINH1在NSCLC中高表达,与肿瘤增殖、转移密切相关,可作为治疗NSCLC的潜在靶点。Objective:To were analyze the expression of Serpin family H member 1(SERPINH1)in non-small cell lung cancer(NSCLC)and its mechanism of action.Methods:Tumor tissues and paracancer tissues of sixty NSCLC patients who underwent tumor resection in Jurong People's Hospital from January 2022 to December 2023 were selected for study.RNAseq data and corresponding clinical information were obtained from the Cancer Genome Atlas(TCGA)dataset for NSCLC tissue and normal tissue.SERPINH1 expression in NSCLC tissues was evaluated by immunohistochemistry.The effects of SERPINH1 on the proliferation,migration and invasion of human lung adenocarcinoma cell line A549 were analyzed in vitro.Results:Compared with normal tissues,SERPINH1 mRNA expression was significantly up-regulated in NSCLC tissues;the overall survival and disease-specific survival of patients with high SERPINH1 expression were shorter than those with low SERPINH1 expression;the SERPINH1 protein expression level in NSCLC tissues was higher than that in paracancer tissues(P<0.001);the proliferation,migration and invasion ability of A549 cells in SERPINH1 overexpression group was higher than that in blank group(P<0.001),and the proliferation,migration and invasion ability of A549 cells in SERPINH1 knockdown group was lower than that in blank group(P<0.001).Conclusion:SERPINH1 is highly expressed in NSCLC,which is closely related to tumor proliferation and metastasis,and can be used as a potential target for the treatment of NSCLC.
