摘 要:Vip3Aa可以进入草地贪夜蛾(Spodoptera frugiperda)的卵巢细胞(Sf9细胞)并诱导凋亡.然而,目前尚不清楚Vip3Aa激活毒素(act Vip3Aa)是否能对昆虫中肠细胞产生同样的作用.本研究发现act Vip3Aa可以进入云杉卷叶蛾(Choristoneura fumiferana)中肠细胞(CF-203细胞)并最终运输到溶酶体中,并且act Vip3Aa进入细胞的数量与毒性呈正相关.act Vip3Aa会引起CF-203细胞溶酶体p H上升、线粒体膜电势下降、基因组DNA断裂、caspase-3活性上升等现象,说明act Vip3Aa能够诱导CF-203细胞凋亡.联合使用多杀菌素(spinosad)和act Vip3Aa能够协同杀伤CF-203细胞,显著提高细胞的死亡率.Vip3Aa can enter the ovary cells of Spodoptera frugiperda(Sf9 cells)and induce apoptosis.However,it is unclear whether Vip3Aa activated toxin(actVip3Aa)exerts the same effects on insect midgut cells.In this study,actVip3Aa was found to be able to internalize into Choristoneura fumiferana midgut cells(CF-203)and fuse with lysosomes,and the internalization level was related to its virulence.Moreover,actVip3Aa treatment could induce increased ly-sosomal pH,decreased mitochondrial membrane potential,genomic DNA fragment,and increased caspase-3 activity.These results suggested that actVip3Aa could induce apoptosis in CF-203 cells.The combined use of spinosad and actVip3Aa significantly increased the mortality of CF-203 cells,implying that spinosad and actVip3Aa had synergistic effects on killing insect cells.
