七氟醚通过PI3K/Akt/eNOS信号通路对大鼠海马树突棘的影响

摘  要:目的探讨吸入七氟醚麻醉对大鼠海马树突棘及其认知功能的影响,并讨论其相关机制。方法48只SD大鼠随机分为对照组,七氟醚-6h组,七氟醚-3h组,七氟醚-1 h组,每组12只。适应性喂养1周,各组大鼠进行水迷宫训练及测试。随后,各组大鼠按照实验方案使用3%七氟醚进行麻醉,吸入七氟醚时间分别为6h、3h和1 h。大鼠恢复清醒24 h后再次进行水迷宫测试,记录大鼠的逃避潜伏时间,穿越平台次数及目标象限巡航时间。试验结束后取海马组织,应用Western blot进行PI3K/Akt/eNOS信号通路相关蛋白检测,利用试剂盒检测Na^(+)-K^(+)-ATP酶活性,运用高尔基染色检测树突棘状态。结果与对照组相比,七氟醚-6h组大鼠逃避潜伏时间显著增加,而PI3K/Akt/eNOS信号通路中PI3K蛋白表达,Akt蛋白与eNOS蛋白磷酸化水平显著降低,Na^(+)-K^(+)-ATP酶活性显著降低,海马树突棘密度和长度都显著降低。结论大鼠过量吸入七氟醚会导致认知功能的损伤,与抑制PI3K/Akt/eNOS信号蛋白的激活,抑制Na^(+)-K^(+)-ATP酶活性的表达,损伤海马树突棘的密度及长度相关。Objective To investigate the effects of anesthesia with sevoflurane on hippocampal dendritic spines and their cognitive function in rats.Methods Forty-eight SD rats were randomly divided into normal control group,3%sevoflurane-6h group,3%sevoflurane-3h group,and 3%sevoflurane-1 h group,with 12 rats in each group.24 hours later,the water maze test was performed,and the escape latency,the number of crossing the platform,and the cruise time in the target quadrant were recorded.The expressions of PI3K/Akt/eNOS signaling pathway-related proteins in hippocampus were detected by Western blot.Na^(+)-K^(+)-ATPase activity was detected by kit,and dendritic spine status was detected by Golgi staining.Results The escape latency was increased,but the expression level of PI3K protein,and Akt and eNOS protein phosphorylation levels were significantly decreased,Na^(+)-K^(+)-ATPase activity reduced,and the density and length of hippocampal dendritic spines was significantly decreased in the sevoflurane-6h group than in the control group.Conclusion Excessive inhalation of sevoflurane leads to cognitive impairment,which is related to inhibiting the activation of related proteins in the PI3K/Akt/eNOS signaling pathway,inhibiting the expression of Na^(+)-K^(+)-ATPase activity and damaging dendritic cells in the hippocampus of rats.
