摘 要:为优化白簕茎总黄酮的提取工艺,比较不同产地白簕茎中总黄酮含量及其抗氧化活性,采用响应面乙醇回流提取法优化白簕茎总黄酮提取工艺,以DPPH和ABTS自由基清除法测定白簕茎总黄酮抗氧化活性。结果表明:白簕茎总黄酮最佳提取工艺为提取时间72 min、乙醇浓度51%、料液比1:40(g/mL);11个产地的白簕茎总黄酮含量差异显著,其中以广东茂名地区的最高,为3.90%,湖南怀化地区的最低,为0.55%;不同地区的白簕茎均具有一定的抗氧化性,其中湖南怀化地区的抗氧化活性最强,对DPPH和ABTS自由基清除率IC_(50)值分别为0.5525、0.5445 mg/mL。综上,优化的提取工艺稳定,适用于各地区白簕茎总黄酮的提取,不同产地提取物具有一定的抗氧化活性,可为其资源开发提供一定的依据。This study aimed to optimize the extraction process of total flavonoids from the stem bark of Acanthopanan trifoliatus and compare the total flavonoid content and antioxidant activity from various regions.The response surface method using ethanol reflux extraction was employed for process optimization,while the antioxidant activity of the stem bark of Acanthopanan trifoliatus was determined using DPPH and ABTS free radical scavenging assays.The results indicated that the optimal extraction conditions were as follows:extraction time of 72 minutes,ethanol volume fraction of 51%,and a liquid-to-solid ratio of 1:40(g/mL).There was a significant variation in total flavonoid content among 11 different regions,with the highest content observed in the Maoming region of Guangdong at 3.90%and the lowest in Huaihua,Hunan at 0.55%.The stem bark of Acanthopanan trifoliatus samples from different regions exhibited notable antioxidant properties,with the highest antioxidant activity observed in the Huaihua region of Hunan,displaying IC_(50) values of 0.5525 mg/mL for DPPH and 0.5445 mg/mL for ABTS free radical scavenging.The optimized extraction process proved to be stable and suitable for extracting total flavonoids from the stem bark of Acanthopanan trifoliatus across various regions.Furthermore,the the stem bark of Acanthopanan trifoliatus extracts from different regions displayed substantial antioxidant activity,providing a valuable foundation for resource development in this field of study.
