摘  要:采用异硫氰酸苯酯(PITC)柱前衍生,建立了红枣中γ-氨基丁酸(GABA)含量的HPLC测定方法,以0.1 mol/L乙酸钠水溶液(pH值6.5,含0.5%三乙胺)和甲醇为流动相进行梯度洗脱,进样量10μL,流速0.7 mL/min,柱温30℃,GABA在0.775~50.000μg/mL内具有良好的线性关系(R^(2)>0.999),检测限(LOD)为0.1467μg/mL,定量限(QOD)为0.4891μg/mL,加样回收率为88.45%~98.13%,在4℃下放置7 d的相对标准偏差RSD仅为4.78%,该方法方便快捷、准确性高、稳定性好,且能够批量分析红枣GABA。不同红枣品种中GABA含量差异显著(p<0.05),其中骏枣的GABA含量最高327.37μg/g DW,酸枣最低为102.22μg/g DW。A HPLC method was established for the determination of gamma-aminobutyric acid(GABA)in jujube by pre-column derivation of phenyl isothiocyanate(PITC).0.1 mol/L sodium acetate aqueous solution(pH value was 6.5,containing 0.5%triethylamine)and methanol were used as mobile phase for gradient elution.The injection volume was 10μL and the flow rate was 0.7 mL/min.The column temperature was 30℃,the GABA had a good linear relationship(R^(2)>0.999)in the range of 0.775~50.000μg/mL,the limit of detection(LOD)was 0.1467μg/mL,the limit of quantification(QOD)was 0.4891μg/mL,and the recoveries were 88.45%~98.13%.The relative standard deviation(RSD)of 7 days at 4℃was only 4.78%.This method was convenient,accurate,stable and can realize the detection method of GABA in batch samples.There were significant differences in GABA content among different jujube varieties(p<0.05),with the highest GABA content of Jun Jujube 327.37μg/g DW and the lowest GABA content of Suanjube jujube 102.22μg/g DW.
