摘 要:氢氧化钙因其价格相对低廉且与其配套的有机酸分离纯化工艺成熟,常作为中和剂应用于有机酸发酵中。但在丁二酸发酵中,因其不能提供丁二酸合成所必需的二氧化碳,存在发酵产量低的问题。文章建立并优化了以氢氧化钙为中和剂结合二氧化碳脉冲补料发酵生产丁二酸的方法。发酵初期不添加中和剂,pH值自然下降至5.7时,立即流加23%氢氧化钙溶液使pH值升至7.5,然后停止添加中和剂,通入0.2 m^(3)/(m^(3)·min)二氧化碳气体至pH值降至6.5时停止通气使发酵液自然发酵,当pH值下降至5.7时,再循环上述操作。使用该方法时,发酵产量为88.47 g/L,糖酸转化率为96.03%,平均生产强度为2.21 g/(L·h),相较于以氢氧化钙为中和剂不补入或者连续通入二氧化碳的方式,糖酸转化率分别提高了298%和95%。该方法为解决使用氢氧化钙作中和剂发酵丁二酸产量低的问题提供了新思路,也为利用现有有机酸分离设备大规模生产丁二酸打下了良好基础。Calcium hydroxide is often used as neutralizing agent in the fermentation of organic acids due to its relatively low price and mature matched organic acid separation and purification process.However,in the fermentation of succinic acid,there is a problem of low fermentation yield due to its inability to provide the necessary carbon dioxide for succinic acid synthesis.In this paper,with calcium hydroxide as the neutralizing agent,combined with carbon dioxide pulse feed fermentation,a method for producing succinic acid is established and optimized.When no neutralizing agent is added in the initial stage of fermentation to naturally lower the pH value to 5.7,23%calcium hydroxide solution is immediately added to raise the pH value to 7.5.Then stop adding neutralizing agent,0.2 m^(3)/(m^(3)·min)carbon dioxide gas is introduced until the pH value drops to 6.5,and stop ventilation to make the fermentation broth naturally ferment.When the pH value drops to 5.7,repeat the above operation.Using this method,the fermentation yield,sugar acid conversion rate,average production intensity are 88.47 g/L,96.03%and 2.21 g/(L·h)respectively.The sugar acid conversion rate increases by 298%and 95%respectively compared to using calcium hydroxide as neutralizing agent, without adding or continuously introducing carbon dioxide. This method has provided a new approach to solve the problem of low yield of succinic acid fermentation using calcium hydroxide as neutralizing agent, and also laid a good foundation for the large-scale production of succinic acid using existing organic acid separation equipment.
