2021-2022年广东省深圳市大鹏新区海水及海产品溶藻弧菌的污染状况、毒力基因分布和耐药性研究

摘  要:目的 调查广东省深圳市海滨旅游区大鹏新区海水及市售海产品中溶藻弧菌污染状况,对溶藻弧菌环境分离株的毒力基因分布特征及耐药性进行分析,为溶藻弧菌感染防控提供数据支持。方法 2021年1月至2022年12月,每季度采集1次大鹏新区近岸海水及市售海产品样本,分离鉴定出溶藻弧菌,用肉汤稀释法测定其对17种抗生素的耐药性;采用聚合酶链式反应方法扩增溶藻弧菌毒力相关基因tdh、trh、tlh、tox R、fur、collagenase、tox S、ure R、fla A、omp W、asp A,产物用1%的琼脂糖凝胶电泳分析。结果 采集海水样本304份,有103份检出溶藻弧菌,检出率为33.88%,不同季节海水中溶藻弧菌的检出率差异有统计学意义(χ^(2)=10.090,P<0.05),第一、第四季度检出率比第二、第三季度要高。采集海产品139份,有108份检出溶藻弧菌,检出率为77.70%。从海水及海产品中共分离鉴定出334株溶藻弧菌,分离的溶藻弧菌对氨苄西林、黏菌素表现出较高的耐药水平,耐药率分别为85.63%、45.94%。11种毒力相关基因均有不同比例的分布。结论 深圳市大鹏新区海水及海产品中溶藻弧菌污染严重,菌株携带的毒力相关基因复杂多样,存在多重耐药,需对环境中溶藻弧菌进行长期监测,包括基因分型、毒力基因分布、耐药性、进化分析等,可为进一步研究溶藻弧菌进化规律及制定防控策略提供数据支持。Objective To investigate the contamination status of Vibrio alginolyticus in seawater and seafood in Dapeng new district of Shenzhen,Guangdong province,analyze the distribution of virulence genes and drug resistance spectrum of V.alginolyticus in the environment and provide data support the control of infections caused by V.alginolyticus.Methods A study was conducted in the Dapeng between January 2021 and December 2022,in which samples of inshore seawater and commercial seafood collected every 3 months were used.V.alginolyticus was isolated and a broth dilution test was used to detect the susceptibilities of the isolates to 17 antibiotics.Polymerase chain reaction was used to amplify V.alginolyticus virulence-related genes including tdh,trh,tlh,toxR,fur,collagenase,toxS,ureR,flaA,ompW,aspA,and the products were analyzed by 1%agarose gel electrophoresis.Results A total of 304 samples of seawater were collected,in which 103 were tested positive for V.alginolyticus,the detection rate was 33.88%.There were significant differences in the detection rate of V.alginolyticus in seawater among different seasons(χ^(2)=10.090,P<0.05),with higher positive rates in the 1st and 4th quarters than in the 2nd and 3rd quarters.A total of 139 samples of seafood were collected,in which 108 were tested positive for V.alginolyticus,the detection rate was 77.70%.A total of 334 strains of V.alginolyticus were isolated from the seawater and seafood samples.The V.alginolyticus isolates showed high levels of resistance to ampicillin and colistin,with resistance rates of 85.63%and 45.94%,respectively.The 11 virulence-related genes were detected with different proportions.Conclusions V.alginolyticus contamination,both in seawater and seafood,was a serious public health problem Dapeng new district.These strains carried complex and diverse virulence-related genes and showed multi-drug resistance.Long-term surveillance for V.alginolyticus in environment,including genotyping,detections of virulence genes and drug resistance and evolutionary anal
