摘 要:目的探究高压氧处理对孤独症幼鼠炎症反应、学习记忆功能及Wnt/β-catenin信号通路的影响。方法将30只受孕成功的雌鼠按照随机数字表法分为正常组(A组)、模型组(B组)和高压氧组(C组),每组10只。B组、C组雌鼠在妊娠12.5 d时腹腔注射600 mg/kg的丙戊酸钠溶液,A组同期腹腔注射同体积生理盐水。B、C组分别产下的幼鼠设定为孤独症幼鼠模型,A组产下的幼鼠为正常对照。然后每组采用随机数字表法再选取10只雄性幼鼠,C组幼鼠给予高压氧处理,A、B组幼鼠每天置于氧舱内,不加压,不吸氧,置入时长与C组相同。采用旷场实验和Morris水迷宫检测各组幼鼠行为学变化;酶联免疫吸附法检测幼鼠血清炎症因子的表达水平;HE染色观察幼鼠海马组织的病理学形态;Western blotting实验检测幼鼠海马组织Wnt/β-catenin信号通路蛋白的相对表达量。结果与B组相比,C组幼鼠旷场中水平穿越格数、直立次数、中央区运动距离明显增多或延长(P<0.05);逃逸潜伏期明显缩短、穿越平台次数明显增多及平台象限停留时间明显延长(P<0.05);海马神经元结构明显改善,炎症细胞浸润明显减轻;血清炎症因子表达水平明显降低(P<0.05)。与A组比较,B组幼鼠海马组织β-catenin蛋白表达明显升高、GSK-3β蛋白表达明显降低(P<0.01);与B组比较,C组幼鼠海马组织β-catenin蛋白表达明显降低、GSK-3β蛋白表达明显升高(P<0.05)。结论高压氧处理孤独症幼鼠通过抑制大脑海马组织Wnt/β-catenin信号通路,有效抑制血清炎症反应,改善和保护神经功能,进一步提高幼鼠的学习记忆能力。ObjectiveTo investigate the effects of hyperbaric oxygen on inflammatory response,learning and memory ability,and Wnt/β-catenin signaling pathway in young rats with autism.MethodsThirty pregnant female rats were randomly divided into normal group(Group A),model group(Group B),and hyperbaric oxygen group(Group C)using the random number table method,with 10 rats in each group.The female rats in groups B and C were intraperitoneally injected with 600 mg/kg sodium valproate solution on day 12.5 of pregnancy,while the rats in Group A were intraperitoneally injected with the same volume of normal saline during the same period.The young rats born in Group B and Group C were set as models of autism,while the young rats born in Group A were set as normal control.Then 10 young male rats were selected from each group,using the random number table method.The young rats in Group C were given hyperbaric oxygen treatment,while the rats in groups A and B were placed in the oxygen chamber every day,without pressurization and oxygen inhalation,but the placement time was the same as that of Group C.The open-field test and Morris water maze were used to detect the behavioral changes of young rats in each group.Enzyme-linked immunosorbent assay was used to detect the expression levels of serum inflammatory factors in young rats.HE staining was used to observe the pathological morphology of hippocampal tissue in the brain of young rats.Western blotting was used to detect the relative expressions of Wnt/β-catenin signaling pathway proteins in the hippocampus of young rats.ResultsCompared with Group B,the horizontal crossings,rearing,and central distance traveled of Group C were significantly increased or extended(P<0.05).Compared with Group B,the escape latency of Group C was significantly shortened,the platform crossings were significantly increased,and the time spent in target quadrant was significantly extended(P<0.05).Compared with Group B,the hippocampal neuronal structure of Group C was significantly improved,and the infiltrati
