摘  要:目的 探讨基质Gla蛋白(MGP)抑制皮肤鳞状细胞癌(cSCC)进展的机制。方法 运用免疫组化学法检测30对cSCC组织与癌旁正常组织中MGP的表达情况,利用癌症基因组图谱数据库对2个独立的SCC数据集(GSE122370和GSE53462)进行MGP基因表达分析;以2株cSCC细胞株(A431、SCL-1)为研究对象,构建MGP过表达稳转细胞系A431、SCL-1细胞株,MTS实验检测cSCC细胞增殖能力;Western blot验证MGP过表达、敲低MGP对cSCC细胞中凋亡相关蛋白的影响。结果 MGP在cSCC组织中表达明显低于癌旁正常组织(P<0.05),并通过对两个在线数据集的分析得到证实。MGP过表达抑制cSCC细胞A431、SCL-1的增殖(P<0.05);MGP过表达后cSCC细胞A431、SCL-1中细胞凋亡的比例明显增加(P<0.05)。上调MGP后A431、SCL-1细胞系中BCL-2表达水平显著下调,Caspase3、Bax的表达则显著上调;敲低MGP后,A431、SCL-1细胞系中BCL-2表达水平显著上调,Caspase3、Bax的表达则显著下调。结论 MGP在cSCC组织中呈低表达,MGP抑制cSCC细胞增殖,同时促进其凋亡;而在机制上,MGP通过调控cSCC细胞中调亡相关蛋白的表达来抑制cSCC细胞增殖并促进癌细胞凋亡,抑制cSCC的发生与发展。Objective To explore the mechanism by which matrix Gla protein(MGP)inhibits the progression of cutaneous squamous cell carcinoma(cSCC).Methods Immunohistochemistry was used to detect the expression of MGP in 30 pairs of cSCC tissues and normal adjacent tissues.MGP gene expression analysis was performed on two independent SCC datasets(GSE122370 and GSE53462)using the Cancer Genome Atlas database.Two cSCC cell lines(A431 and SCL-1)were used as research objects to construct MGP overexpression stable transgenic cell lines A431 and SCL-1,and MTS assay was used to detect the proliferation ability of cSCC cells.The effects of MGP overexpression and knockdown on apoptosis-related proteins in cSCC cells were verified by Western blot.Results The expression of MGP in cSCC tissue was significantly lower than that in adjacent normal tissue(P<0.05),which was confirmed by analysis of two online datasets.MGP overexpression inhibited the proliferation of cSCC cells A431 and SCL-1(P<0.05).After overexpression of MGP,the proportion of apoptosis in cSCC cells A431 and SCL-1 significantly increased(P<0.05).After upregulating MGP,the expression level of BCL-2 was significantly downregulated in A431 and SCL-1 cell lines,while the expression of Caspase3 and Bax was significantly upregulated.After knocking down MGP,the expression level of BCL-2 was significantly upregulated in A431 and SCL-1 cell lines,while the expression of Caspase3 and Bax was significantly downregulated.Conclusion MGP shows low expression in cSCC tissues,which inhibits cell proliferation and promotes apoptosis;in terms of mechanism,MGP inhibits the proliferation of cSCC cells,promotes the apoptosis of cancer cells,and inhibits the occurrence and development of cSCC by regulating the expression of middle note apoptosis related proteins in cSCC cells.
