摘  要:目的:探讨MAP3K9通过调控Notch-1促进胰腺癌细胞增殖、迁移的机制。方法:在胰腺癌细胞中分别转染MAP3K9过表达质粒和siRNA,RT-qPCR检测MAP3K9 mRNA表达水平,MTT实验检测转染后细胞增殖能力的改变,流式细胞术检测细胞凋亡能力的变化,划痕试验检测细胞迁移能力的变化,Transwell实验检测细胞的迁移与侵袭能力,Western Blot检测MAP3K9、Notch-1及EMT相关蛋白的表达情况。结果:RT-qPCR和Western Blot显示,PaTu-8988和PANC-1细胞的MAP3K9表达水平低于BXPC-3细胞;RT-qPCR结果表明,转染MAP3K9过表达质粒可以显著提高PaTu-8988和PANC-1细胞MAP3K9 mRNA表达水平,而转染siRNA MAP3K9可明显下调BXPC-3细胞MAP3K9 mRNA表达水平;上调胰腺癌细胞MAP3K9表达,可提高胰腺癌细胞增殖、迁移与侵袭能力,并抑制细胞凋亡;在胰腺癌细胞中过表达MAP3K9,可上调Notch-1、Vimentin、N-cadherin以及slug蛋白表达水平,并降低E-cadherin表达水平。结论:MAP3K9能够激活Notch-1,进而促进胰腺癌细胞的上皮—间充质转化(EMT)过程,增强胰腺癌细胞增殖及迁移能力。Objective:To clarify the mechanism of MAP3K9 promoting the proliferation and migration of pancreatic cancer cells by regulating Notch-1.Methods:MAP3K9 overexpression plasmid and siRNA were transfected into pancreatic cancer cells respectively.The expression level of MAP3K9 mRNA was detected by RT-qPCR,the cell proliferation ability after transfection was detected by MTT assay,flow cytometry was used to detect the cell apoptosis,wound healing assay was used to measure the change of cell migration capacity,the migration and invasion ability of cells was detected by Transwell assay,and the expression of MAP3K9,Notch-1 and EMT related proteins was detected by Western Blot.Results:RT-qPCR and Western Blot showed that the expression levels of MAP3K9 in PaTu-8988 and PANC-1 cells were lower than those in BXPC-3 cells.The RT-qPCR results identified that transfection of MAP3K9 overexpressing plasmid significantly increased the mRNA expression level of MAP3K9 in PaTu-8988 and PANC-1 cells.while transfection of siRNA MAP3K9 significantly downregulated the mRNA expression level of MAP3K9 in BXPC-3 cells.Upregulation of MAP3K9 expression in pancreatic cancer cells enhanced the proliferation,migration and invasion capacity of pancreatic cancer cells,but suppressed cell apoptosis ability.Consistently,overexpression of MAP3K9 in pancreatic cancer cells can increase the expression of Notch-1,Vimentin,N-cadherin and slug proteins,while reduce the expression of E-cadherin.Conclusion:MAP3K9 promotes the epithelial-mesenchymal transtition process and enhances the capacity of cell proliferation and migration of pancreatic cancer through upregulating Notch-1.
