摘 要:目的建立紫归方中紫草及当归的含量测定方法,并优选其提取工艺。方法以高效液相色谱波长切换联合梯度洗脱法同时测定紫归方中阿魏酸和左旋紫草素的含量。以液料比、提取溶剂、提取时间为考察因素,分别以左旋紫草素和阿魏酸含量为评价指标,采用L9(3^(4))正交实验设计,优选紫草和当归的提取工艺并验证。结果左旋紫草素在5.15~206 ng(R^(2)=0.9997)范围内与吸收度呈良好的线性关系,平均加样回收率为110.5%,RSD为1.1%。阿魏酸在5.13~205.2 ng(R^(2)=0.9999)范围内与吸收度呈良好的线性关系,平均加样回收率为110.5%,RSD为1.1%。最佳提取工艺为:液料比1:35,75%乙醇作为提取溶剂,超声提取40分钟,出膏率约为36.5%。结论优选的工艺和方法简便可行,可用于紫归方中紫草及当归的质量控制和提取。Objective To establish a method for determining the contents of ferulic acid and shikonin in Zigui Formula(紫归方)and optimize the extraction process of Zigui Formula.Methods The contents of ferulic acid and shikonin in Zigui Formula were determined by HPLC wavelength switching combined with gradient elution method.The extraction process was optimized by orthogonal experimental design of L_(9)(3^(4)),with the ratio of liquid to solid,extraction solvent and extraction time as the factors,and the contents of ferulic acid and shikonin as the evaluation indexes.Results The method had a good linearity at 5.13-205.2 ng(R^(2)=0.9999)for ferulic acid and 5.15-206 ng(R^(2)=0.9997)for shikonin.The recoveries were 111.2%and 110.5%,respectively and the values of RSD were 1.2%and 1.1%,respectively.The optimal extraction process of Zigui Formula was as follows:the liquid-solid ratio was 1:35;75%alcohol was used as the extraction solvent;the ultrasonic extraction lasted for 40 minutes;the paste yield was about 36.5%.Conclusion The optimized process and method are simple and feasible,and can be used for the quality control and extraction of Zigui Formula.
