摘 要:目的 采用分子分类方法鉴定2023年10月重庆市万州区发生的三起食用野生蘑菇中毒事件相关样品,为突发中毒事件蘑菇鉴定及预防蘑菇中毒提供参考。方法 提取蘑菇样品基因组DNA,分别扩增ITS、LSU片段,扩增产物克隆测序,综合序列信息的Gen Bank数据库BLAST分析、FN数据库检索和种系发生结果,确定蘑菇品种。结果 三起事件的6份蘑菇样品均成功扩增ITS、LSU基因片段并克隆测序,ITS和LSU基因序列Gen Bank数据库BLAST分析,ITS基因序列FN数据库检索,分别属于粉褶菌属(Entoloma)和牛肝菌属(Suillus),但是品种名称存在差异。进一步结合ITS序列种系发生结果,综合判定三起蘑菇中毒事件中,事件1样品(WZ1S)和事件3的样品(WZ4S、WZ5S、WZ6S)为波缘粉褶蕈(Entoloma sinuatum),事件2样品WZ2S和WZ3S分别为粘盖乳牛肝菌(Suillus bovinus)和琉珀乳牛肝菌(Suillus placidus),3个品种均为毒蘑菇。结论 分子分类方法应用于蘑菇中毒事件调查,能够快速确定毒蘑菇种类,有助于提升实验室突发中毒事件调查鉴定能力和预防蘑菇中毒。Objective To investigate the samples related to three mushroom poisoning incidents that occurred in Wanzhou District,Chongqing in October 2023 using molecular taxonomy methods,providing a reference for mushroom identification and prevention of mushroom poisoning in emergency situations.Methods Genomic DNA was extracted from mushroom sam-ples,and ITS and LSU fragments were amplified,cloned,and sequenced.The sequence information was analyzed through BLAST in the GenBank database,searched in the FN database,and phylogenetic results were integrated to determine the mushroom species.Results Six mushroom samples from the three incidents successfully amplified the ITS and LSU gene fragments,which were cloned and sequenced.BLAST analysis of the ITS and LSU gene sequences in the GenBank database,along with FN database searches,revealed that the samples belonged to the genera Entoloma and Suillus,although there were discrepancies in species names.Further phylogenetic analysis of the ITS sequences determined that samples from Incident 1(WZ1S)and samples from Incident 3(WZ4S,WZ5S,WZ6S)were identified as Entoloma sinuatum,while samples WZ2S and WZ3S from Incident 2 were identified as Suillus bovinus and Suillus placidus,respectively.All three species are toxic mush-rooms.Conclusion The application of molecular taxonomy methods in mushroom poisoning incident investigations allows for rapid identification of toxic mushroom species,enhancing laboratory capabilities for investigating and preventing mushroom poisoning incidents.
