胃癌组织中RUNX3蛋白表达及其与幽门螺杆菌感染的相关性分析

摘　　要：目的探讨胃癌组织中RUNX家族转录因子3(RUNX family transcription factor 3,RUNX3)蛋白表达及其与幽门螺杆菌(HP)感染的相关性。方法收集2020年1月至2023年1月在浙江省乐清市人民医院接受治疗的90例胃癌患者(胃癌组)、80例胃癌癌前病变患者(癌前病变组)、100例慢性浅表性胃炎患者(慢性浅表性胃炎组)的病灶组织,采用免疫组化检测RUNX3蛋白表达情况,免疫印迹法检测RUNX3蛋白相对表达量,实时荧光定量PCR(RT-qPCR)检测RUNX3 mRNA相对表达量,快速尿素酶试验和改良Ciemsa法检测HP感染情况。结果与慢性浅表性胃炎组比较,癌前病变组的RUNX3蛋白表达阳性率(41.25%比64.00%)、RUNX3蛋白相对表达量[(0.54±0.06)比(1.12±0.12)]、RUNX3 mRNA相对表达量[(1.64±0.14)比(2.35±0.24)]明显降低(P<0.01)。与癌前病变组比较,胃癌组的RUNX3蛋白表达阳性率(16.67%比41.25%)、RUNX3蛋白相对表达量[(0.21±0.04)比(0.54±0.06)]、RUNX3 mRNA相对表达量[(1.00±0.02)比(1.64±0.14)]明显降低(P<0.01)。与HP阴性的病灶组织比较,胃癌组(12.68%比31.58%)、癌前病变组(26.47%比52.17%)及慢性浅表性胃炎组(40.91%比70.51%)HP阳性的病灶组织RUNX3蛋白阳性表达率均明显降低(P<0.01)。胃癌组织(r=-0.683)、癌前病变组织(r=-0.582)及慢性浅表性胃炎组织(r=-0.701)的RUNX3蛋白相对表达量均与HP阳性率呈负相关(P<0.01)。结论HP可能通过下调RUNX3蛋白表达参与胃癌的发生发展。Objective To investigate the expression of RUNX family transcription factor 3(RUNX3)protein in gastric cancer tissue and its correlation with Helicobacter pylori(HP)infection.Methods Lesion tissues were collected from 90 patients with gastric cancer,80 patients with precancerous lesions,and 100 patients with chronic superficial gastritis,who received treatment at our hospital from January 2020 to January 2023.The expression of RUNX3 protein was detected using immunohistochemistry.Western blotting was used to detect the relative expression of RUNX3 protein.Real-time fluorescence quantitative PCR was used to detect the expression of RUNX3 mRNA.HP infection was detected using a rapid urease test and the modified Ciemsa method.Results Compared to the chronic superficial gastritis group,the precancerous lesion group had a significantly lower positive rate of RUNX3 protein expression(41.25%vs.64.00%),reduced RUNX3 protein level[(0.54±0.06)vs.(1.12±0.12)],and decreased RUNX3 mRNA level[(1.64±0.14)vs.(2.35±0.24)](P<0.01).Compared to the precancerous lesion group,the gastric cancer group had a significantly lower positive rate of RUNX3 protein expression(16.67%vs.41.25%),reduced RUNX3 protein level[(0.21±0.04)vs.(0.54±0.06)],and decreased RUNX3 mRNA level[(1.00±0.02)vs.(1.64±0.14)](P<0.01).Compared to HP-negative tissues,the expression rates of RUNX3 in the gastric cancer group(12.68%vs.31.58%),the precancerous lesion group(26.47%vs.52.17%),and the chronic superficial gastritis group(40.91%vs.70.51%)with HP positive were significantly lower(P<0.01).The relative expression levels of RUNX3 protein in gastric cancer tissues(r=-0.683),precancerous lesions(r=-0.582),and chronic superficial gastritis tissues(r=-0.701)were negatively correlated with HP positivity(P<0.01).Conclusion HP infection may be involved in the occurrence and development of gastric cancer by down-regulating the expression of RUNX3 protein.
