摘 要:本文建立了一种应用超高效液相色谱-串联质谱仪同时测定畜禽肉中肾上腺素类、拟肾上腺素类、肾上腺素代谢物共10种肾上腺素类药物的检测方法。考察了不同前处理条件和仪器条件对检测结果的影响,从而选出最佳解决方案。目标物以乙腈提取,分散固相萃取法(QuEChERS)净化,用Shim-pack GIST C18作为色谱柱,甲醇和含0.1%甲酸的5 mmol·L^(-1)乙酸铵水溶液作为流动相,UPLC-MS/MS确证检测,外标法定量。该方法定量限0.2~2.0μg·kg^(-1),回收率75.6~107.5%,相对标准偏差1.1~8.7%。该方法与传统检测方法相比,具有检测药物种类多、最低检测限低、稳定性好、简便快捷、节省试剂等优势。同时应用该方法对市售的畜禽肉样本进行检测分析,确定多种肾上腺素类药物在畜禽肉中的残留情况,为更好地控制市场上的畜禽肉品质提供技术支撑。A method was established for simultaneous detection of 10 adrenaline related drugs composed of adrenaline,pseudoadren-aline,and adrenaline metabolites in livestock and poultry meat using ultra high performance liquid chromatography tandem mass spectrometry(UPLC-MS/MS).The effects of dfferent pre-treatment conditions and instrument conditions on the detection results were investigated,in order to select the best way.Acetonitrile was used to extract the substances we need to detect from the sample,and then QuEChERS was used to purify the substances we need to detect,the Shim pack GIST C18 was used to be chromatographic column,the instrument used for the detection,methanol and 5 mmol·L^(-1)ammonium acetate aqueous solution containing O.1%for-mic acid was used as mobile phases,and quantification of this method was UPLC-MS/MS,detected and calibrated the target we need to detect through specific outternal standard.The quantitative limit of this method was 0.2~2.0μg·kg^(-1),recovery rate75.6~107.5%,relative standard deviation 1.1~8.87%.Compared with traditional detection methods,it had advantages such as a wide range of drug types,low minimum detection limit,good stability,simplicity and efficiency,and saves reagents.At the same time,this method was applied to detect and analyze samples of commercially available livestock and poultry meat,to determine the residual situation of various adrenaline drugs in livestock and poultry meat,providing technical support for better control of the quality of livestock and poultry meat in the market.
