摘 要:目的观察督脉点刺法对缺血缺氧脑损害大鼠的记忆功能和细胞坏死的作用。方法7日龄的SD幼鼠6窝共40只,并随机分成空白组,假手术组,模型组,针刺组,造模成功2 d后,运用督脉点刺法对针刺组进行治疗,其余组别大鼠相同条件进行饲养,治疗1个疗程(10 d)后,对幼鼠进行水迷宫检查,在测试完毕后采用免疫组化法检查各组大鼠海马区Bax,Bcl-2,Caspase-3阳性表达结果。结果空白组、假手术组和针刺组的水迷宫实验正确次数多于模型组(P<0.05);与空白组、假手术组比较,模型组、针刺组幼鼠海马区的Bax、Caspase-3蛋白表达水平更高,Bcl-2蛋白表达水平降低(均P<0.05);与模型组比较,针刺组幼鼠海马区Bax、Caspase-3蛋白表达水平更低,Bcl-2蛋白表达升高(P<0.05)。结论督脉点刺法可以改善缺血缺氧脑损伤大鼠的学习记忆能力,其机制与Bax,Bcl-2,Caspase-3细胞凋亡相关。Objective To observe the effects of acupoint acupuncture on learning and memory ability and cell apoptosis in rats with ischemic and hypoxic brain injury.Methods A total of 6 litters of 7-day-old sd rats,A total of 40,were randomly divided into blank group,sham operation group,model group and acupuncture group.2 days after successful modeling,acupuncture group was treated with the method of acupoint acupuncture,and the other groups were fed under the same conditions.The water maze test was performed on the young rats.After the test,the positive expression of Bax,Bcl-2and Caspase-3 in the hippocampus of the rats in each group was detected by immunohistochemistry.Results The correct times of water maze test in the blank group,Sham Operation Group and acupuncture group were more than those in the model group(P<0.05),the expression levels of Bax and Caspase-3 in hippocampus of model group and acupuncture group were higher than those of model group,and the expression levels of Bcl-2 were lower than those of model group(P<0.05),the expression levels of Bax and Caspase-3 in hippocampus were lower and the expression of BCL-2 was higher in acupuncture group(P<0.05).Conclusions The acupoint acupuncture method can improve the learning and memory ability of rats with ischemic and hypoxic brain injury,and its mechanism is related to the apoptosis of Bax,Bcl-2 and Caspase-3 cells.
