四翼滨藜叶绿体基因组组装及序列特征分析

摘  要:本文以库布齐沙漠边缘的四翼滨藜为实验材料,采用CTAB法提取其全基因组DNA,利用Illumina双端测序技术获得了完整的四翼滨藜叶绿体基因组序列,对四翼滨藜叶绿体基因组进行组装、注释及序列特征分析,并构建系统发育树。结果表明:四翼滨藜叶绿体基因组全长为15064 bp,共注释出132个基因,含有87个编码蛋白基因,8个rRNA基因和37个tRNA基因;MISA检测到四翼滨藜有49个SSR位点,为单核苷酸重复和二核苷酸重复型,碱基组成以A/T碱基类型为主;四翼滨藜一共含有20698个密码子,其中编码亮氨酸的密码子数量最多,存在30个偏好密码子,有29个以A/U碱基结尾;系统发育树表明四翼滨藜与中亚滨藜有较密切的亲缘关系。本文对四翼滨藜叶绿体基因组的研究将为苋科植物的物种鉴定、遗传进化分析和DNA条形码的构建奠定基础。In this study, Atriplex canescens from the edge of Kubuqi Desert was used as the experimental material.The whole-genome DNA was extracted using the CTAB method, and the complete chloroplast genome sequence of Atriplex canescens was obtained using the Illumina paired-end sequencing technology.The chloroplast genome of Atriplex canescens was then assembled, annotated, and its sequence characteristics were analyzed.Additionally, a phylogenetic tree was constructed.The results revealed that the chloroplast genome of Atriplex canescens has a total length of 150664 bp and contains 132 annotated genes, including 87 protein-coding genes, 8 rRNA genes, and 37 tRNA genes.A total of 49 SSR sites were detected in Atriplex canescens using MISA,mainly consisting of mononucleotide and dinucleotide repeats, with A/T as the major base composition.The chloroplast genome of Atriplex canescens contains a total of 20698 codons, with leucine-coding codons being the most abundant classes.Thirty preferred codons were identified, with 29 ending in A/U.The phylogenetic tree analysis indicated a close relationship between Atriplex canescens and Atriplex centralasiatica.The study of the chloroplast genome of Atriplex canescens will lay a foundation for species identification, genetic evolution analysis, and DNA barcode construction in Amaranthaceae plants.
