摘 要:目的:探讨吲哚胺2,3–双加氧酶1(IDO1)在肝癌与癌旁组织中的表达情况及其作用。方法:应用公共数据库获取IDO1在泛癌和肝癌中的表达数据。应用免疫荧光染色法对82例肝癌和配对癌旁组织以及14例无配对的肝癌组织中IDO1的表达情况进行检测。皮下接种人源肝癌细胞SK–HEP1建立裸鼠皮下移植瘤模型,并予以IDO1抑制剂干预,通过苏木精–伊红(HE)病理染色和增殖细胞核抗原(PCNA)免疫组化染色评价IDO1抑制剂对肝癌细胞在体内生长速度的影响,免疫荧光染色检测瘤体组织中簇分化抗原(CD)4^(+)T细胞和CD8^(+)T细胞的浸润情况。结果:IDO1在多种癌组织中高表达。与癌旁组织相比,肝癌组织中IDO1的表达水平明显升高。病理染色结果示,抑制IDO1的表达可以明显降低肝癌移植瘤组织中PCNA的表达水平,增加CD4^(+)T细胞和CD8^(+)T细胞的浸润。结论:IDO1在肝癌组织中高表达,下调IDO1的表达可抑制肝癌细胞在体内增殖,延缓皮下瘤体的生长速率,增强免疫细胞对肿瘤细胞的应答。Objective To investigate the expression and role of indoleamine 2,3-dioxygenase 1(IDO1)in liver cancer and adjacent tissues.Methods The expression data of IDO1 in pancarcinoma and hepatocellular carcinoma were obtained by applying public databases.Immunofluorescence staining was used to detect IDO1 expression level in 82 cases of hepatocellular carcinoma,paired adjacent tissues and 14 cases of unpaired hepatocellular carcinoma.Subcutaneous transplantation tumor model of nude mice was established by subcutaneous inoculation of human hepatocellular carcinoma cells SK-HEP1,and IDO1 inhibitor was applied to intervene.Effects of IDO1 inhibitor on the growth rate of hepatocellular carcinoma cells in vivo was evaluated by hematoxylin–eosin(HE)pathological staining and proliferating cell nuclear antigen(PCNA)immunohistochemical staining.The infiltration of cluster of differentiation(CD)4^(+)T cells and CD8^(+)T cells in tumor tissues was detected by immunofluorescence staining.Results IDO1 was highly expressed in various carcinoma tissues.The expression level of IDO1 in hepatocellular carcinoma tissues was significantly higher than that in paracancer tissues.Pathological staining results showed that inhibition of IDO1 expression could significantly reduce the expression level of PCNA and increase the infiltration of CD4^(+)T cells and CD8^(+)T cells in liver cancer transplanted tumor tissues.Conclusion IDO1 is highly expressed in liver cancer tissues,and down-regulation of IDO1 expression can inhibit the proliferation of liver cancer cells in vivo,delay the growth rate of subcutaneous tumors,and enhance the response of immune cells to tumor cells.
