摘  要:目的 探讨尼古丁刺激对人鼻咽癌5-8F细胞增殖、分化的影响。方法 培育人鼻咽癌5-8F细胞,用0~20μmol/L的尼古丁处理5-8F细胞,通过CCK-8法检测尼古丁对于细胞活力的影响。针对经尼古丁刺激后鼻咽癌细胞体外培养的细胞侵袭和细胞迁移能力的变化,则分别采用划痕和Transwell试验。为进一步探求其背后可能潜在的机制,试验选取了PI3K/AKT/mTOR、CDK1/CCNB1/PLK1等经典信号通路,通过q-RCR试验检测与通路密切相关的上下游分子的调控基因,如细胞周期依赖性蛋白激酶抑制因子(p21)、周期蛋白依赖性激酶1(CDK1)、波形蛋白基因(VIM)、细胞转移相关基因(Snail2)等的表达水平。结果 不同浓度(0~20μmol/L)的尼古丁处理鼻咽癌5-8F细胞后,细胞活性呈先升高后下降趋势。尼古丁组(2~10μmol/L)同对照组(0)比较,差异有统计学意义(P <0.01),提示尼古丁可以促进鼻咽癌5-8F细胞增殖,并对5-8F细胞的迁移和侵袭能力均具有促进作用(P <0.05)。另外,尼古丁可以促进5-8F细胞的CDK1、VIM基因表达上调,p21基因表达下调(P <0.05)。结论 在鼻咽癌细胞的体外培养试验中,经尼古丁刺激作用后的5-8F细胞的增殖、迁移和侵袭能力均得到一定程度的增强。此外,尼古丁能够促进肿瘤细胞分化,阻滞肿瘤细胞消亡,可能是导致鼻咽癌发生、发展的潜在关键危险因素。Objective To investigate the effect of nicotine stimulation on the proliferation and differentiation of human nasopharyngeal carcinoma(NPC) 5-8F cells.Methods 5-8F cells were cultured and treated with different concentrations(0-20 μmol/L) of nicotine.Then the effect of nicotine on cell viability was detected by CCK-8 method.Scratch and Transwell experiments were used to observe the changes of cell invasion and cell migration ability of nasopharyngeal carcinoma cells cultured in vitro after nicotine stimulation.In order to further explore the potential mechanism behind it,the classic signaling pathways such as PI3K/AKT/mTOR,CDK1/CCNB1/PLK1 were selected.The expression levels of regulatory genes of upstream and downstream molecules closely related to the pathway,such as cyclin-dependent protein kinase inhibitor(p21),cyclin-dependent kinase 1(CDK1),vimentin gene(VIM),cell metastasis-related gene(Snail2) and so on,were detected by q-RCR assay.Results After treatment of nasopharyngeal carcinoma 5-8F cells with different concentrations(0-20 μmol/L) of nicotine,the cell activity increased first and then decreased.The difference between the nicotine group(2-10 μmol/L) and the control group(0) was statistically significant(P<0.01),suggesting that nicotine can promote the proliferation of nasopharyngeal carcinoma 5-8F cells,and promote the migration and invasion of 5-8F cells(P<0.05).On the other hand,nicotine could up-regulate the expression of CDK1 and VIM genes and down-regulate the expression of p21 gene in 5-8F cells(P<0.05).Conclusions In the in vitro culture experiment of nasopharyngeal carcinoma cells,the proliferation,migration and invasion ability of 5-8F cells stimulated by nicotine were enhanced to a certain extent.In addition,nicotine can promote tumor cell differentiation and block the extinction of tumor cells,which may be a potential key risk factor for the occurrence and development of nasopharyngeal carcinoma.
