摘 要:铜、铁超载与多种肝脏疾病发生发展关系密切。铜死亡是一种铜依赖的新型细胞死亡方式,细胞内铜离子异常聚集与三羧酸循环中的脂酰化成分直接结合,进而导致线粒体脂化蛋白聚集与铁硫蛋白簇蛋白下调,引起蛋白质毒性应激导致细胞死亡。铁死亡是一种铁依赖的新型细胞死亡方式,主要特征是活性氧和脂质过氧化物累积。本文对铜、铁超载及其引发的铜、铁死亡在肝脏疾病发生发展中的作用机制进行综述,旨在为相关疾病的防治提供理论依据。Copper and iron overload are closely related to the occurrence and development of various liver diseases.Copper death is a new type of copper-dependent cell death,in which the abnormal accumulation of intracellular copper ions directly combines with the fatty acyl components of the tricarboxylic acid cycle,which leads to the accumulation of mitochondrial fatty proteins and the down-regulation of iron-sulfur protein cluster proteins,resulting in proteotoxic stress and cell death.Iron death is a new type of irondependent cell death characterized by the accumulation of reactive oxygen species and lipid peroxides.In this paper,the mechanism of copper and iron overload and the death caused by copper and iron in the occurrence and development of liver diseases was reviewed in order to provide theoretical basis for the prevention and treatment of related diseases.
