摘  要:目的观察劳力型热射病大鼠肝损伤的病理变化特点及核因子E2相关因子2(Nrf2)/血红素加氧酶(HO)-1的表达情况。方法选用健康雄性SD大鼠,随机分为空白组(Con组)、劳力型热射病组(构建劳力型热射病大鼠动物模型,分为EHS 2 h组、EHS 6 h组和EHS 12 h组)共4组,每组6只大鼠。所有大鼠均经过梯度强度适应性跑台后开始实验。Con组大鼠在经过适应性跑台训练后正常饲养。劳力型热射病组进行跑台训练直至大鼠发生劳力型热射病。按照发病后2、6、12 h随机选取EHS组大鼠,并留取肝组织备用。采用苏木精伊红染色观察肝形态学,利用免疫组织化学染色法检测大鼠肝Nrf2、磷酸化核因子E2相关因子2(pNrf2)、HO-1蛋白表达情况。结果与正常组比较,热射病各组肝细胞肿胀、胞浆脂肪变性及肝细胞坏死明显,门静脉淤血、扩张。免疫组织化学染色结果显示:Nrf2、pNrf2、HO-1在EHS组不同时间点阳性细胞表达均高于正常组,差异有统计学意义(P<0.05)。结论Nrf2/HO-1信号通路在劳力型热射病大鼠肝损伤中存在保护作用。Objective To observe the pathological changes of liver injury and the expression of Nrf2/HO-1 in exertional heat stroke rats.Methods Healthy male SD rats were randomly divided into blank group(Con group)and exertional hyperthermia group(EHS 2 h group,EHS 6 h group and EHS 12 h group)with 6 rats in each group.The experiment was started after all rats underwent gradient intensity adaptive running.Con group rats were fed normally after adaptive treadmill training.The exertional heat stroke group was given treadmill training until the rats developed exertional heat stroke.EHS group rats were randomly selected 2 h,6 h and 12 h after the onset of the disease,and liver tissue was reserved for use.The liver morphology was observed by HE staining.The expression of Nrf2,pNrf2 and HO-1 proteins in the liver of rats was detected by immunohistochemical staining.Results Compared with the normal group,the hepatocyte swelling,cytoplasmic steatosis and hepatocyte necrosis were obvious,and the portal vein was congestion and dilatation.Immunohistochemical staining showed that the expression of Nrf2,pNrf2 and HO-1 in positive cells in EHS group was significantly different from that in normal group at different time points(P<0.05).Conclusion Nrf2/HO-1 signaling pathway has protective effect on liver injury in exertional heat stroke rats.
