两种金福菇的蛋白鉴定及金属元素含量的测定

摘  要:为深入研究和开发金福菇Tg505、大峡谷的营养价值和功能,对其中含有的蛋白及金属元素进行测定和分析。采用饱和酚抽提法和二维电泳技术,获得金福菇Tg505、大峡谷的蛋白指纹图谱,并筛选出其中的高表达蛋白;再利用基质辅助激光解析电离飞行时间质谱仪鉴定高表达蛋白的酶水解产物及确定蛋白名称,并通过生物信息学手段对蛋白功能进行初步分析;利用电感耦合等离子体质谱技术对两种金福菇金属元素含量进行测定。结果表明,金福菇Tg505及大峡谷均有33个高表达蛋白,涉及金福菇生长、发育、遗传、抗逆、应激及药物活性等多种功能,两种金福菇中含量较高的金属元素依次为K、Mg、Mo、Ca、Al、Fe。本研究不仅能够对进一步深入开发、利用具有药食同源特性的金福菇提供指导,而且对海南热带食用菌的研究和开发具有参考意义。To deeply explore and develop their nutritive values and functions,the proteins and metal elements in two kinds of Macrocybe lobayensis(ML),TG505 and Grand Canyon ML were determined and analyzed.Protein fingerprints were obtained using phenol extraction and two-dimensional electrophoresis(2DE),and highly expressed proteins were selected.The enzymatic hydrolysates of highly expressed proteins were identified by matrix-assisted laser desorption ionization timeof-flight mass spectrometry(MALDI-TOF-MS).Functional analysis of these proteins was carried out using bioinformatics tools.The contents of metal elements in the two kinds of mushrooms were determined by inductively coupled plasma mass spectrometry(ICP-MS).The results showed that 33 highly expressed proteins were identified from each kind of ML,which were involved in the growth,development,genetics,stress resistance,stress response,pharmaceutical activity and other functions of ML.For both kinds of ML,the major metal elements were K,Mg,Mo,Ca,Al,and Fe in decreasing order.The present study not only provides guidance for further exploitation and utilization of ML,with both medicinal and culinary uses,but also is of reference significance for the research and development of tropical edible mushrooms in Hainan.
