摘  要:目的探索氨基胍(anminoguanidine,AG)对胶质瘤瘤周(2 mm)脑水肿的影响。方法将90只雄性Wistar大鼠随机分为5组,每组18只,分别为:假手术组(A组)、模型对照组(B组)、模型+AG 75 mg/kg组(C组)、模型+AG 150 mg/kg组(D组)、模型+AG 225 mg/kg组(E组)。B、C、D、E组大鼠脑右侧尾状核注射10μl的C6细胞建立胶质瘤模型;A组按上述方法注射10μl培养基。14 d后模型建立,A、B组给予对应剂量生理盐水(ip;bid),C、D、E组给予对应剂量AG(ip;bid),3 d。干湿重法测脑含水量;免疫荧光和Western-blot法检测瘤周目的蛋白闭合蛋白1(ZO-1)、闭锁蛋白(Occludin)、血管内皮生长因子(VEGF)、水通道蛋白4(AQP4)的含量。结果与B组相比,D组能显著降低脑含水量(P<0.05),C、E组没有显著降低脑含水量(P>0.05)。与B组相比,D组显著增加瘤周脑组织ZO-1、Occludin蛋白表达(P<0.05),并显著降低VEGF、AQP4蛋白表达(P<0.05)。结论AG通过提高瘤周脑组织ZO-1和Occludin表达、降低VEGF和AQP4表达,减轻胶质瘤脑水肿。Objective To explore the effects of aminoguanidine(AG)on brain edema around glioma(2 mm).Methods 90 male Wistar rats were randomly divided into 5 groups with 18 rats in each group:sham operation group(A),model control group(B),model+AG 75 mg/kg group(C),model+AG 150 mg/kg group(D)and model+AG 225 mg/kg group(E).The glioma model was established by injecting 10μl C6 cells into the right caudate nucleus of Wistar rats in group B,C,D and E,whereas group A was injected 10μl medium according to the above method.14 days later,the model was established.Group A and B were administered corresponding dose of normal saline(ip;bid),while group C,D and E were administered corresponding dose of AG(ip;bid)for 3 days.The brain water content was measured by dry-wet weight method,and the contents of Zonula Occluden1(ZO-1),Occludin,vascular endothelial growth factor(VEGF)and aquaporin 4(AQP4)were detected by immunofluorescence and Western-blot methods.Results Compared with group B,group D significantly decreased the brain water content(P<0.05),while group C and E did not significantly decrease the brain water content(P>0.05).Compared with group B,group D significantly increased the expression of ZO-1 and Occludin protein(P<0.05),and significantly decreased the expression of VEGF and AQP4 protein in the peritumoral brain tissue(P<0.05).Conclusions AG can alleviate the brain edema caused by glioma by increasing the expression of ZO-1 and Occludin and reducing the expression of AQP4 and VEGF in peritumor brain tissue.
