基于网络药理学与分子对接探讨益气聪明汤治疗颈性眩晕作用机制

摘  要:目的基于网络药理学和分子对接研究益气聪明汤治疗颈性眩晕(CV)的作用机制。方法利用TCMSP数据库筛选益气聪明汤的活性成分及其相关作用靶点,通过GeneCards、OMIM数据库获取CV靶点,将获得的益气聪明汤作用靶点与CV疾病靶点取交集,导入STRING数据库进行分析,并利用Cytoscape3.7.2软件构建蛋白相互作用(PPI)网络,利用R4.2.6软件进行GO功能及KEGG通路富集分析,利用AutoDock1.5.7软件进行分子对接并将其可视化。结果共检索出益气聪明汤活性成分207个、作用靶点256个。核心活性成分包括槲皮素、山柰酚、β-谷甾醇、豆甾醇、刺芒柄花素,与CV相关靶点3230个,其中以AKT1、IL6、TP53、CASP3、VEGFA等为核心靶点。KEGG通路富集分析显示,治疗CV主要涉及AGE-RAGE、IL-17和TNF等信号通路。分子对接结果显示,核心靶点与核心活性成分对接活性较强。结论益气聪明汤可能通过槲皮素、山柰酚、β-谷甾醇、豆甾醇、刺芒柄花素等活性成分作用于AKT1、IL6、TP53、CASP3和VEGFA等靶点,调控AGE-RAGE、IL-17和TNF等信号通路发挥治疗CV的作用。Objective To explore the mechanism of Yiqi Congming Decoction in treating cervical vertigo(CV)based on network pharmacology and molecular docking technique.Methods TCMSP was used to screen out the active components and related action targets of Yiqi Congming Decoction.CV-related targets were obtained through GeneCards and OMIM databases,and the obtained action targets of Yiqi Congming Decoction were intersected with CV-related disease targets and imported into the STRING database for analysis.In addition,Cytoscape 3.7.2 software was used to construct the protein interaction(PPI)network.R4.2.6 software was used for GO function and KEGG pathway enrichment analysis,and AutoDock 1.5.7 software was used for molecular docking and visualization.Results A total of 232 kinds of active components and 256 targets of Yiqi Congming Decoction were retrieved.The core active components included quercetin,kaempferol,beta-sitosterol,stigmasterol,and formononetin,and 3230 CV-related targets were identified,among which AKT1,IL6,TP53,CASP3,VEGFA and so on were the core targets.KEGG pathway enrichment analysis showed that AGE-RAGE,IL-17 and TNF signaling pathways were mainly involved in the treatment of CV.The results of molecular docking showed that the core target had strong docking activity with the core active components.Conclusion Yiqi Congming Decoction may act on AKT1,IL6,TP53,CASP3 and VEGFA through active components such as quercetin,kaempferol,beta-sitosterol,stigmasterol,and formononetin,to regulate AGE-RAGE,IL-17 and TNF signaling pathways in treating CV.
