动物双歧杆菌乳亚种XLTG11对抗生素扰乱小鼠肠道菌群调节作用研究

摘  要:通过设计抗生素混合溶液(硫酸庆大霉素7.5 mg/次+头孢噻肟钠30 mg/次)扰乱小鼠肠道菌群模型,探究动物双歧杆菌乳亚种XLTG11调节小鼠肠道菌群的作用。将60只BALB/c小鼠随机分成5组,每组12只。分别为阴性对照组、模型对照组、低剂量组、中剂量组和高剂量组,小鼠连续14 d每天灌胃抗生素混合溶液(硫酸庆大霉素7.5 mg/次+头孢噻肟钠30 mg/次)诱导肠道菌群失调模型。然后,各剂量组分别灌胃动物双歧杆菌乳亚种XLTG11。检测小鼠体质量变化、肠道菌群变化、进行肠道组织病理学分析,检测肠道屏障,检测血清中脂多糖(LPS)和D-乳酸含量,检测短链脂肪酸含量。结果表明,高剂量的动物双歧杆菌乳亚种XLTG11能够显著降低模型小鼠结肠组织损伤程度,显著降低血清中LPS和D-乳酸含量,显著上调ZO-1、Occludin、Claudin-1和MUC2相关基因的表达水平、增加短链脂肪酸含量。调节肠道菌群组成,小鼠粪便中双歧杆菌和乳杆菌含量明显增加,高剂量动物双歧杆菌乳亚种XLTG11可以使肠杆菌、肠球菌和产气荚膜梭菌含量显著减少。动物双歧杆菌乳亚种XLTG11可改善模型小鼠肠道菌群组成,提高短链脂肪酸含量,提高肠道屏障相关基因表达,进而有效调节抗生素相关性肠道菌群失调。This study was to explore the effect of Bifidobacterium animalis subsp.lactis XLTG11 on the regulation of gut microbiota in mice by designing an antibiotic mixed solution(Gentamicin sulfate 7.5 mg/time+Cefotaxime sodium 30 mg/time)to disturb the mouse gut microbiota model.60 BALB/c mice were randomly divided into 5 groups,with 12 mice in each group.The mice were divided into negative control group,model control group,low-dose group,medium dose group and high-dose group.The mice were fed with antibiotic mixed solution(Gentamicin sulfate 7.5 mg/time+Cefotaxime sodium 30 mg/time)every day for 14 consecutive days to induce the gut microbiota imbalance model.Subsequently,each dose group was gavaged with Bifidobacterium animal subsp.lactis XLTG11.The body weight and gut microbiota of mice were measured,the intestinal histopathological analysis was carried out,the intestinal barrier was detected,the content of Lipopolysaccharide(LPS)and D-lactic acid in serum was detected,and the content of short-chain fatty acid was detected.Results:showed that high dose of XLTG11 of Bifidobacterium lactis could significantly reduce the degree of colon tissue injury in model mice,significantly reduce the content of LPS and D-lactic acid in serum,significantly increase the expression level of ZO-1,Occludin,Claudin-1 and MUC2 related genes,and increase the content of short-chain fatty acid.By regulating the composition of gut microbiota,the contents of bifidobacteria and lactobacillus in mouse feces were significantly increased,and the contents of enterobacteria,Enterococcus and Clostridium perfringens were significantly reduced by high dose of Bifidobacterium animal subsp.lactis XLTG11.Bifidobacterium animal subsp.lactis XLTG11 can improve the composition of gut microbiota in model mice,increase the content of short-chain fatty acid,increase the expression of intestinal barrier related genes,and effectively regulate the imbalance of antibiotic related gut microbiota.
