摘 要:目的:基于临床生物信息学探讨白藜芦醇对自闭症谱系障碍(ASD)的潜在治疗作用。方法:从基因表达综合数据库(GEO)中获取白藜芦醇和ASD的全基因组表达谱数据;分别对白藜芦醇和ASD的全基因组表达谱数据进行差异基因分析、基因本体(GO)富集分析、京都基因与基因组百科全书(KEGG)通路富集分析和蛋白质相互作用网络(PPI)分析并筛选核心基因;采用领域知识得分法在中英文数据库中对白藜芦醇具有重要调控作用核心基因的有效性进行文本验证。结果:白藜芦醇组和ASD组分别筛选出294个和256个差异基因(DEGs);白藜芦醇组GO富集的基因主要与神经系统发育、神经发生、突触组织等有关,KEGG通路富集主要与动物线粒体自噬、胆固醇代谢、甲状腺激素合成等有关;ASD组KEGG通路富集主要与丝裂原活化蛋白激酶信号通路、甲状腺激素合成、缩宫素信号通路等有关;白藜芦醇组DEGs的PPI网络分析中共获得11个核心基因,主要与细胞周期、突触发生和突触可塑性、炎症、免疫反应等有关。结论:白藜芦醇具有抑制炎症反应、促进免疫调节、改善氧化应激、调节突触发生和突触生成过程、调节神经递质的释放等功能,对ASD具有潜在的治疗意义。Objective:To explore the potential therapeutic effect of resveratrol on autism spectrum disorders(ASD)based on clinical bioinformatics.Methods:Obtain whole genome expression profiles of resveratrol and ASD from GEO database;Perform differentially expressed gene analysis,Gene Ontology enrichment analysis,Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis,and protein-protein interaction network analysis on the whole genome expression profile data of resveratrol and ASD,and screen for core genes;Using domain knowledge scoring method to validate the effectiveness of core genes with important regulatory effects on resveratrol in Chinese and English databases.Results:294 and 256 differentially expressed genes(DEGs)were identified in the resveratrol group and ASD group,respectively;The genes enriched in the resveratrol group GO are mainly related to nervous system development,neurogenesis,synaptic tissue,etc.The KEGG pathway enrichment is mainly related to animal mitochondrial autophagy,cholesterol metabolism,thyroid hormone synthesis,etc;The enrichment of KEGG pathway in ASD group is mainly related to mitogen activated protein kinase signaling pathway,thyroid hormone synthesis,oxytocin signaling pathway,etc;A total of 11 core genes were identified in the PPI network of resveratrol group DEGs,mainly related to cell cycle,synaptogenesis and synaptic plasticity,inflammation,and immune response,etc.Conclusions:Resveratrol has functions such as inhibiting inflammatory response,promoting immune regulation,improving oxidative stress,regulating synaptogenesis and synaptogenesis processes,and regulating neurotransmitter release.It has potential therapeutic significance for ASD.
