摘  要:为探索提高牡丹皮芍药苷提取水平的新途径,开展了UDP-糖基转移酶产生菌筛选及其对牡丹皮发酵的研究。从酱醅中筛选获得一株UDP-糖基转移酶产生菌GT16,经分子学鉴定为德巴利酵母(Debaryomyces prosopidis)。通过摇瓶发酵实验,确定了该菌种发酵的最佳温度和最佳初始pH分别为28℃和6.5。表面活性剂Triton X-100促进牡丹皮前体发酵转化为芍药苷效果明显,其最佳添加量为0.04%,最佳的发酵时间为72 h。研究表明,GT16菌种对牡丹皮的发酵转化作用明显,发酵样品的芍药苷浓度是未发酵对照样品的123.6%。Triton X-100对该菌种发酵的促进作用明显,添加Triton X-100的发酵样品的芍药苷浓度是无添加对照样品的115.9%。In order to explore a new approach to improve the extraction level of paeoniflorin from peony bark,the screening of UDP-glycosyltransferase producing bacteria and its effect on the fermentation of peony bark were carried out.An UDP-glycosyltransferase producing strain GT16 was isolated from fermented paste and identified as Debaryomyces prosopidis.The optimal fermentation temperature and initial pH of the strain GT16 were 28℃ and 6.5,respectively.Furthermore,the optimal addition amount of Triton X-100 to promote the fermentation of peony bark precursor into paeonidin was 0.04%,and the optimal fermentation time was 72 h.The fermentation and transformation effect of the strain GT16 on peony bark was significant.The concentration of paeoniflorin in the fermented sample was 123.6% of that in the unfermented control sample.Triton X-100 obviously promoted the fermentation of the strain GT16,and the concentration of paeoniflorin in the fermented sample added with Triton X-100 was 115.9% of that in the control sample without addition.
