摘　　要：目的:探讨MDA-9在结肠癌细胞中的表达及对SW620细胞迁移和侵袭的影响。方法:RT-qPCR和Western blot分别检测人正常结肠上皮细胞NCM460和3种结肠癌细胞(HCT116、SW480、SW620)中MDA-9的mRNA和蛋白表达水平。在SW620细胞中转染MDA-9靶向干扰序列(si-MDA-9)及对照序列(si-NC),划痕实验检测细胞迁移能力,Transwell实验检测细胞侵袭能力,Western blot检测p-FAK、MMP-2蛋白水平的变化。结果:3种结肠癌细胞中MDA-9的mRNA表达水平(P<0.001)及蛋白表达水平(P<0.01)均高于NCM460细胞。si-MDA-9组SW620细胞相对迁移率(60.7±5.8)%及侵袭率(55.3±10.3)%,均低于si-NC组的(98.3±14.3)%和(98.4±9.5)%,及空白对照组的(100±9.8)%和(100±13.1)%,差异具有统计学意义(P<0.01)。且si-MDA-9组细胞中p-FAK(P<0.05)和MMP-2(P<0.01)蛋白水平也均低于si-NC组和空白对照组。结论:MDA-9在结肠癌细胞中高表达,抑制其表达水平将导致SW620细胞迁移和侵袭能力下降,这可能和下调p-FAK、MMP-2蛋白表达相关。Objective:To detect the expression of MDA-9 in colorectal cancer cells,and determine its role on migration and invasion of SW620 cells.Methods:The mRNA and protein expression levels of MDA-9 in normal colorectal epithelial cell line(NCM460)and three colorectal cancer cell lines(HCT116,SW480 and SW620)were determined by RT-qPCR and Western blot assay.The small interfering RNA targeting MDA-9(si-MDA-9)and negative control(si-NC)were transfected into SW620 cells,and scratch healing assay and Transwell assay were used to detect the effect on cell migration and invasion.The proteins expression of p-FAK and MMP-2 was determined by Western blot assay.Results:MDA-9 mRNA expression level(P<0.001)and protein expression level(P<0.01)were significantly up-regulated in three colorectal cancer cell lines than in NCM460 cells.The relative migration ability and invasion ability of SW620 cells in the si-MDA-9 group were(60.7±5.8)%and(55.3±10.3)%,which were lower than(98.3±14.3)%and(98.4±9.5)%in the si-NC group,and(100±9.8)%and(100±13.1)%in the blank control group(P<0.01).The protein levels of p-FAK(P<0.05)and MMP-2(P<0.01)in SW620 cells of si-MDA-9 group were also lower than those of si-NC group and blank control group.Conclusion:MDA-9 is highly expressed in colorectal cancer cells,and inhibiting its expression level will lead to a decline in the migration and invasion ability of SW620 cells,which may be related to inhibiting the expression of p-FAK and MMP-2 proteins.
