摘  要:目的研究小鼠在全肠外营养(total parenteral nutrition,TPN)模型下与正常小鼠相比,肠道中真菌群落的多样性以及组成结构的改变。方法建立TPN小鼠模型后,采集TPN模型小鼠(n=5)和正常对照小鼠(n=5)的新鲜粪便样本,利用ITS DNA测序技术对肠道真菌进行测定,然后通过生物信息学方法对两组小鼠的真菌菌群多样性、群落结构及功能特征的差异进行分析。结果TPN模型小鼠和正常对照小鼠的肠道真菌Alpha多样性具有明显的差异(P<0.05);Beta多样性存在统计学差异(P<0.01);在TPN模型小鼠中,Candida(假丝酵母属)的相对丰度明显减少(P<0.01),Penicillium(青霉菌属)的相对丰度明显减少(P<0.01),Aspergillus(曲霉菌属)的相对丰度明显减少(P<0.01),Talaromyces(篮状菌属)的相对丰度明显减少(P<0.01)。LEfSe分析揭示,在正常对照小鼠中,Candida(假丝酵母属),Penicillium(青霉菌属),Aspergillus(曲霉菌属)和Talaromyces(篮状菌属)有明显的富集。结论与正常小鼠相比,TPN模型小鼠的肠道真菌菌群组织结构发生明显改变。肠道真菌菌群失调可能促进TPN相关并发症的发生,调节肠道真菌群落平衡可能成为预防TPN相关并发症的一种新策略。Objective To analyze the diversity and composition changes of gut fungal communities between mouse model of total parental nutrition(TPN)and normal control mice.Methods After mouse model of TPN was constructed,fresh feces were collected from TPN mice(n=5)and normal control mice(n=5).Internal transcribed spacer(ITS)DNA sequencing was applied to determine intestinal fungi,and then bioinformatics analysis was conducted to identify the differences in fungal diversity,structure,and functional properties between the 2 groups of mice.Results There were significant differences in Alpha diversity(P<0.05)and Beta diversity(P<0.01)of intestinal fungi between the 2 groups.In the TPN model group,the relative abundances of Candida,Penicillium,Aspergillus and Talaromyces were obviously reduced(all P<0.01).LEfSe analysis indicated that the above 4 strains were notably enriched in the normal control mice.Conclusion TPN mice exhibit characteristic changes in the composition of gut fungal flora compared to normal control mice.Dysfunction of gut fungal community may promote the occurrence of TPN related complications,and regulating the balance of gut fungal community may become a new strategy for preventing TPN related complications.
