摘  要:通过Tris辅助法合成粒径均匀的胶体金颗粒,用于建立一种快速、方便的甲萘威胶体金免疫层析检测方法。优化试纸条的p H值、抗体使用量,并将胶体金标记抗体用于胶体金试纸条组装,测验试纸条的灵敏度、特异性、准确性、重复性、稳定性。结果显示:研制的甲萘威胶体金试纸条检测限为0.60 mg/kg,符合国家对甲萘威最大残留量的要求,与灭多威、仲丁基、异丙威、丁基呋喃、涕灭威等相似结构化合物不产生反应,且4个批次生产的试纸条检测结果均一致,在常温干燥条件下可保存10个月。通过目测判定结果,该方法简便,快速,适合现场快速检测。This experiment synthesized colloidal gold nanoparticles with uniform particle size using the Tris-assisted method to establish a rapid and convenient colloidal gold immunochromatographic assay for the detection of carbaryl.The pH value of the test strips and the amount of antibody used were optimized,and colloidal gold labeled antibody was prepared and assembled into colloidal gold test strips.The sensitivity,specificity,accuracy,repeatability,and stability of the test strips were evaluated.The results showed that the detection limit of the carbaryl colloidal gold test strips developed in this experiment is 0.6 mg/kg,which meets the national requirements for the maximum residue limit of carbaryl and showed no react with similar structurally compounds such as methomyl,sec-butyl,isoprocarb,butylfuran,and aldicarb.The test results of the four batches of production were consistent,and the test strips can be stored for 10 months under normal dry conditions.The method is simple,fast,and suitable for on-site rapid detection.
