摘  要:为建立完善的紫菀组织培养与快速繁殖体系,解决种苗退化问题,以采自亳州的紫菀为试验材料,开展植物组织培养技术研究,筛选出紫菀外植体最佳消毒时间和紫菀最佳增殖培养基、生根培养基,并观察紫菀组培苗炼苗移栽后的生长状况。结果表明:用0.1%升汞处理8 min,紫菀外植体污染与褐化水平较低,成活率为46.67%;最佳继代增殖培养基为MS+6-BA 1.0 mg/L+NAA 0.2 mg/L,增殖系数为4.56;最佳生根培养基为MS+IBA 0.3 mg/L+NAA 0.5 mg/L,生根率为96.67%;紫菀组培苗移栽60 d的生长状况优于移栽1、30 d,叶片数较多,叶片宽较宽,叶片长和叶柄长均较长。In order to establish a comprehensive tissue culture and rapid propagation system for Aster tataricus and solve the problem of seedling degradation,this paper studied the plant tissue culture technology by using Aster tataricus collected from Bozhou as experimental materials,screened the optimal disinfection time for Aster tataricus explant,selected the best proliferation culture medium and rooting culture medium for Aster tataricus,and observed the growth of Aster tataricus tissue culture seedlings after seedling refinement and transplantation.The results showed that the pollution and browning level of Aster tataricus explant treated with 0.1%mercuric chloride for 8 minutes were low,with a survival rate of 46.67%;the optimal subculture medium for proliferation was MS+6-BA 1.0 mg/L+NAA 0.2 mg/L,with a proliferation coefficient of 4.56;the optimal rooting medium was MS+IBA 0.3 mg/L+NAA 0.5 mg/L,with a rooting rate of 96.67%;the growth of Aster tataricus tissue culture seedlings transplanted for 60 days was better than that transplanted for 1,30 days,with more leaves,wider leaf width,longer leaf length and petiole length.
