基于孟德尔随机化分析胃食管反流病与牙龈疼痛的关系

摘　　要：目的通过采用两样本孟德尔随机化(MR)分析,研究胃食管反流病与牙龈疼痛之间的因果关系。方法从已发表的全基因组关联研究中获得与牙龈疼痛相关的单核苷酸多态性,从已发表的全基因组关联研究中获得与胃食管反流病相关的汇总水平遗传数据,研究主要使用逆方差加权法(IVW)进行两样本MR分析。使用Cochran中Q统计学方法来评估结果的异质性,同时采用MR-Egger检测水平多效性,“留一法”进行敏感性分析。结果胃食管反流病对牙龈疼痛呈现正因果效应(OR=1.009,95%CI:1.006~1.012,P<0.001)。结论胃食管反流病的遗传性增加与发生牙龈疼痛风险的增加显著相关。深入理解胃食管反流病和牙龈疼痛之间的正相关关系对于胃食管反流病的临床治疗和患者的口腔健康管理具有重要意义。Objective To investigate the causal relationship between gastroesophageal reflux disease(GERD)and gingival pain by using two-sample Mendelian randomization(MR)analysis.Methods Single nucleotide polymorphisms associated with gingival pain were obtained from published genome-wide association studies,and poled level genetic data associated with GERD were obtained from published genome-wide association studies.Two Benmure randomizations were performed using inverse variance weighting(IVW).The Q statistical method in Cochran was used to evaluate the heterogeneity of the results,and the MR-Egger was used to detect the level of pleiotropy,and the sensitivity analysis was carried out by"leave-one-out"analysis.Results GERD had a positive causal effect on gingival pain(OR=1.009,95%CI:1.006-1.012,P<0.001).Conclusion Increased heritability of GERD is significantly associated with increased risk of gingival pain.A thorough understanding of the positive correlation between GERD and gingival pain is of great significance for clinical treatment of GERD and oral health management of patients.
