摘　　要：结直肠癌(colorectal cancer,CRC)是消化系统中最常见的恶性肿瘤类型之一。CRC的发病率逐年上升,也是癌症相关死亡的第二大常见原因。大多数CRC患者有结直肠息肉病变突变,而剩余1/2的患者有β-连环蛋白(β-catenin)基因突变。并且与癌变相关的Wnt/β-连环蛋白信号通路在CRC中被异常激活。所以靶向Wnt/β-连环蛋白信号通路治疗CRC具有临床价值。目前为止,随着对Wnt/β-连环蛋白信号通路的深入研究,已经开发出了能够抑制或阻碍CRC发展的抑制剂。本文综述β-catenin、Wnt/β-连环蛋白信号通路在结直肠息肉和CRC中的作用,概述Wnt/β-连环蛋白信号通路抑制剂的研究现状,并提出相关抑制该通路的潜在靶点。Colorectal cancer(CRC)is one of the most common malignant tumors in the digestive system.The incidence of CRC is increasing year by year,CRC is also the second most common cause of cancer-related deaths.The majority of CRC patients have colorectal polyposis mutations,while the remaining half have mutations in theβ-catenin gene.And the Wnt/β-catenin signaling pathway associated with carcinogenesis is abnormally activated in CRC.Therefore,targeting Wnt/β-catenin signaling pathway has clinical value in the treatment of CRC.So far,with the in-depth study of the Wnt/β-catenin signaling pathway,inhibitors that can inhibit or hinder CRC development have been developed.In this review,the roles ofβ-catenin and Wnt/β-catenin signaling pathway in colorectal polyps and CRC are reviewed,the research status of inhibitors of Wnt/β-catenin signaling pathway is reviewed,the potential targets to inhibit this pathway are proposed.
