摘  要:已有研究表明伪狂犬经典疫苗株Bartha-K61对小鼠具有较强的致死性,为了研究伪狂犬疫苗株Bartha-K61对兔的致病性,本研究以10^(3)TCID_(50)的剂量接种5只成年家兔,家兔于接种后第3 d出现死亡,并陆续在5 d内全部死亡。对病死兔进行解剖后发现大脑血管扩张及出血,心脏出现坏死以及肺部气肿、出血、坏死等典型的伪狂犬病病理变化。病理切片也显示心脏血管扩张,血管周围轻度水肿,脑部胶质细胞增生,细胞数量增多,其他各脏器也都有不同程度的病理变化。通过荧光定量PCR的方法对病死兔各组织的病毒载量进行了测定。结果表明,病死兔的各组织脏器均检测出高水平的病毒DNA,均显著高于对照组。为了进一步研究伪狂犬疫苗株Bartha-K61对家兔的致病性,本研究测定了Bartha-K61对家兔的半死致死量,结果表明:Bartha-K61对家兔的半数致死量为10^(0.3)TCID_(50),以1×TCID_(50)的剂量接种成年家兔后,仍可引起20%的家兔致死,由此可见Bartha-K61对家兔的致死性要显著强于小鼠。因此在使用Bartha-K61过程中应针对不同物种注意生物安全控制。Many studies have shown that the classical pseudorabies vaccine strain Bartha-K61 has a strong lethal eff ect on mice.In order to explore the pathogenicity of the pseudorabies vaccine strain Bartha-K61 in rabbits,5 adult rabbits were inoculated with 10^(3)TCID_(50)in this study.The rabbits started to die on the third day after inoculation and all of them died successively within 5 days.Necropsy of the dead rabbits showed typical lesions of pseudorabies such as cerebral vascular dilation and hemorrhage,heart necrosis and pulmonary emphysema,hemorrhage and necrosis.Pathological sections also showed vascular dilatation of hearts,mild perivascular edema,proliferation and increase of glial cells in brains and some changes of diff erent degrees in other organs.The viral loads of dead rabbit tissues were determined by fl uorescence quantitative PCR.The results showed that high levels of viral DNA were detected in the tissues and organs of dead rabbits.In order to further study the pathogenicity of pseudorabies vaccine strain Bartha-K61 in rabbits,the lethal dose of Bartha-K61 was measured in this study.The median lethal dose of Bartha-K61 to rabbits was 10^(0.3)TCID_(50).After inoculation with 1×TCID_(50),Bartha-K61 caused death in 20%of adult rabbits.In conclusion,the lethal eff ect of Bartha-K61 on rabbits was signifi cantly stronger than that on mice.Therefore,attention should be paid to biosafety control of Bartha-K61 use in diff erent animal species.
