摘 要:琼脂糖是一种从石花菜、江蓠等红藻中提取的线性多糖,在生物、医药等领域具有广泛应用。但目前国内琼脂糖的制备工艺较为复杂、成本较高且各批次琼脂糖的质量稳定性较差,使得国内科研机构及生物、医药行业过度依赖进口琼脂糖。利用过氧化氢辅助聚乙二醇法可对琼脂进行绿色脱硫与对制备的琼脂糖进行提纯,通过单因素实验优化得出过氧化氢脱硫的最佳条件为:无水乙醇浓度70%,过氧化氢添加量15 mL,反应时间2 h,反应pH 9,反应温度50℃。在此条件下制备得到过氧化氢改性琼脂的硫酸根含量为0.21%、凝胶强度为966.6 g/cm^(2)。再用聚乙二醇法对过氧化氢改性的琼脂进行提纯,制备的琼脂糖的硫酸根含量为0.19%,凝胶强度为1244.9 g/cm^(2)。开发的过氧化氢辅助聚乙二醇法是一种简单、快速、高效、低成本、环保的琼脂糖制备方法,将有助于解决国内琼脂糖“卡脖子”的问题。Agarose is a linear polysaccharide extracted from red algae such as Gelidium and Gracilaria,and it has widespread applications in biology,pharmaceuticals,and other fields.However,the domestic preparation process for agarose is complex,costly,and characterized by poor quality stability across different batches,leading to an excessive reliance on imported agarose by domestic research institutions and the biopharmaceutical industry.The use of hydrogen peroxide-assisted polyethylene glycol method can greenly desulfurize agar and purify the prepared agarose.Through single-factor experiments,the optimal desulfurization conditions using hydrogen peroxide were determined to be:anhydrous ethanol concentration at 70%,hydrogen peroxide addition amount at 15 mL,reaction time of 2 h,reaction pH at 9,and reaction temperature at 50℃.Under these conditions,the sulfate content of hydrogen peroxide-modified agar was approximately 0.21%,with the gel strength of 966.6 g/cm^(2).Furthermore,the purification of the hydrogen peroxide-modified agar using the polyethylene glycol method resulted in agarose with a sulfate content of 0.19%and a gel strength of 1244.9 g/cm^(2).The developed hydrogen peroxide-assisted polyethylene glycol method is a simple,rapid,efficient,low-cost,and environmentally friendly method for agarose preparation,which will help solve the“bottleneck”issue of domestic agarose production.
