摘 要:为研究临床监测中分离两株H9N2亚型禽流感病毒A/chicken/Henan/F1108/2019及A/chicken/Shandong/F07/2021跨种感染哺乳动物能力,试验构建了H9基因系统发生树,采用红细胞吸附试验及固相结合试验测定两株病毒受体结合特性,并通过小鼠感染试验探究两株病毒对小鼠感染性及致病性。结果显示:A/chicken/Henan/F1108/2019及A/chicken/Shandong/F07/2021与近些年人及禽分离毒株进化关系较近;A/chicken/Henan/F1108/2019主要结合α-2,3唾液酸,而A/chicken/Shandong/F07/2021主要结合α-2,6唾液酸;A/chicken/Henan/F1108/2019在小鼠鼻甲及肺脏中的复制能力及致病性均强于A/chicken/Shandong/F07/2021。研究表明具有α-2,3唾液酸受体结合特性的H9N2亚型禽流感病毒在鸡群中仍然存在,且分离株对小鼠具有较强的致病性。In order to study the ability of two strains of H9N2 subtype avian influenza virus(A/chicken/Henan/F1108/2019 and A/chicken/Shandong/F07/2021)isolated by our laboratory from infect mammals,a phylogenetic tree of H9 gene was constructed,erythrocyte adsorption assay and solid binding assay were used to determine the receptor binding characteristics of the two strains,and finally the infectivity and pathogenicity of the two strains of viruses to mice were explored by mouse infection test.The results showed that A/chicken/Henan/F1108/2019 and A/chicken/Shandong/F07/2021 were closely related to the evolution of human and poultry isolates in recent years.The A/chicken/Henan/F1108/2019 strain mainly bindedα-2,3 sialic acid,while the A/chicken/Shandong/F07/2021 strain mainly bindedα-2,6 sialic acid.The replication ability and pathogenicity of A/chicken/Henan/F1108/2019 in the turbinates and lungs of mice were stronger than those of A/chicken/Shandong/F07/2021.The results indicated that the H9N2 subtype avian influenza virus withα-2,3 sialic acid receptor binding properties still existed in chickens,and the isolate had strong pathogenicity to mice.
