2株H9N2亚型禽流感病毒的分离鉴定及遗传变异分析

摘  要:为深入了解禽流感病毒(Avian influenza virus,AIV)分子生物学特征,试验对河南省某发病蛋鸡场疑似AIV感染的病鸡组织进行病原分离,并对分离病原进行全基因组测序以及遗传进化和序列分析。结果显示:共分离2株H9N2亚型AIV,分别为A/chicken/Henan/HN22/2022(H9N2)和A/chicken/Shangqiu/01/2021(H9N2);2株分离株HA基因均位于h9.4.2.5谱系的分支2,内部基因与来自不同宿主的H7N9、H3N8等亚型AIV具有高度同源性且亲缘关系较近;2株分离株的HA蛋白裂解位点是PSRSSR,与低致病性AIV特性一致,其受体结合位点左侧臂发生Q234L突变,NA蛋白颈部第62~64位存在氨基酸缺失;接种2株分离株的试验鸡没有出现明显临床症状,没有导致气管和肺脏出血,与该亚型其他分离株不同。研究表明,试验分离的2株H9N2亚型AIV关键位点突变情况相对稳定,但仍有一些适应性突变产生,可能出现多种亚型同时感染鸡的情况。In order to understand the molecular biology of avian influenza virus(AIV),the pathogen was isolated from chicken tissue suspected to be infected with AIV in an infected layer farm in Henan province,and the whole genome sequencing,genetic evolution and sequence analysis of the isolated pathogen were performed.The results showed that two strains of H9N2 subtype AIV were isolated,named as A/chicken/Henan/HN22/2022(H9N2)and A/chicken/Shangqiu/01/2021(H9N2),respectively.The HA genes of the two isolates were located on branch 2 of the h9.4.2.5 pedigree,and the internal genes of which were highly homologous to H7N9 and H3N8 subtype AIV from different hosts.The cleavage site of the HA protein of the two isolates was PSRSSR,which was consistent with the characteristics of low pathogenic AIV.The Q234L mutation in the left arm of the receptor binding site;And there was a deletion of amino acids in the neck of the NA protein at amio acid position of 62-64.Chicken infected with the two isolates showed no obvious clinical signs and did not cause tracheal or lung bleeding,which was different from other isolates of this subtype.The study suggested that the mutational profile at key sites of the two isolates of H9N2 subtype AIV was relatively stable,but there still occurred some adaptive mutations,which might be the possibility of co-infection of chickens with multiple subtypes.
