摘 要:目的探讨干扰素诱导蛋白5(IFIT5)在肺癌组织中的表达及可能的分子机制。方法采用免疫组织化学染色法检测IFIT5在85例正常癌旁组织及85例肺癌癌组织中的表达。采用了慢病毒转染法,分别下调和上调IFIT5在A549和NCI-H1703细胞中的表达,并通过CCK8法、Transwell实验、划痕实验检测干扰前后A549和NCI-H1703细胞增殖、迁移和侵袭能力变化。通过qRT-PCR和Westernblot法检测干预前后相关mRNA及蛋白的表达变化。结果IFIT5在肺癌组织中表达水平显著高于相应癌旁组织(P<0.05);敲低IFIT5可抑制肺癌细胞A549和NCI-H1703的增殖、迁移和侵袭能力,上皮间质转化(EMT)相关蛋白E-cadherin蛋白的表达增高,而N-cadherin和Vimentin蛋白表达减少(P<0.05);相反的,过表达IFIT5可促进肺癌细胞A549和NCI-H1703的增殖、迁移和侵袭能力,EMT相关蛋白E-cadherin蛋白的表达降低,而N-cadherin和Vimentin蛋白表达增高(P<0.05)。体内实验表明敲低IFIT5对体内肺癌细胞具有显着的抑制作用。结论IFIT5在肺癌组织中表达升高,高表达IFIT5能够通过调控A549和NCI-H1703细胞EMT促进其侵袭和迁移。Objective The study aims to explore the expression of interferon-induced protein 5(IFIT5)in lung cancer tis‐sues and its possible molecular mechanism.Methods Lentiviral transfection was used to down-regulate and up-regulate the expres‐sion of IFIT5 in A549 and NCI-H1703 cells,respectively.CCK8 assay,Transwell assay and scratch assay were used to detect the changes in the proliferation,migration and invasion of A549 and NCI-H1703 cells before and after the intervention.qRT-PCR and Western blot were used to detect the changes in the expression of related mRNA and protein before and after the intervention.Results The expression level of IFIT5 in lung cancer tissues was significantly higher than that in corresponding para-cancerous tissues(P<0.05).Knocking down IFIT5 may inhibit the proliferation,migration and invasion of lung cancer cells A549 and NCI-H1703.The expres‐sion of epithelial-to-mesenchymal transition(EMT)-related protein E-cadherin increases,while the expression of N-cadherin and Vimentin decreases(P<0.05).On the contrary,overexpression of IFIT5 may promote the proliferation,migration and invasion of lung cancer cells A549 and NCI-H1703.The expression of EMT-related protein E-cad‐herin protein is reduced,while the expression of N-cadherin and Vi‐mentin protein is increased(P<0.05).In vivo experiments show that knocking down IFIT5 has a significant inhibitory effect on lung cancer cells in vivo.Conclusion LIFIT5 expression is elevated in lung cancer tissues,and high expression of IFIT5 can promote the invasion and migration of A549 and NCI-H1703 cells by regulating EMT.
