摘  要:为了探究葡萄糖促进解淀粉芽孢杆菌(Bacillus amyloliquefaciens)BB61合成纳米硒(SeNPs)的机制,研究了不同浓度葡萄糖对菌体生长、SeNPs合成以及相关基因表达水平的影响。结果表明:在亚硒酸钠胁迫下,B.amyloliquefaciens BB61的nfsA、trxA和trxB基因表达水平极显著上调(P<0.01),且具有显著剂量效应(P<0.05),namA基因表达水平虽也显著上调,但剂量效应并不显著;此外,亚硒酸钠的出现和浓度升高还会导致tcyC、tcyA、gabD、zwf、nhaC、fdhF、KSO-RS02125和KSO-RS17170等基因的表达水平显著上调,这些基因分别参与亚硒酸盐的转运、糖代谢、电子链传递和SeNPs的分泌。以0.5 g/L亚硒酸钠为底物时,LB培养基中添加20 g/L葡萄糖对促进菌体蛋白和SeNPs的合成最佳,72 h时SeNPs转化率为72.33%(对照为53.25%),上述基因表达水平均显著上调。综上,硫氧还蛋白还原系统(TrxA、TrxB)和亚硝酸还原酶(NfsA)可能对于高浓度亚硒酸钠环境下SeNPs的转化更为关键;LB培养基中适当添加葡萄糖可显著提高SeNPs合成过程中各关键酶基因的表达水平和SeNPs的转化效率。To investigate the mechanism of glucose promoting the synthesis of selenium nanoparticles(SeNPs)by Bacillus amyloliquefaciens BB61,the effects of different concentrations of glucose on bacterial growth,SeNP synthesis,and related gene expression levels were studied.The results showed that under sodium selenite stress,the expression levels of nfsA,trxA and trxB genes in B.amyloliquefaciens BB61 were extremely significantly upregulated(P<0.01)and had a significant dose effect(P<0.05);the expression level of namA gene was significantly upregulated,but the dose effect was not significant.In addition,the presence and increased concentration of sodium selenite could significantly upregulate the expression levels of genes such as tcyC,tcyA,gabD,zwf,nhaC,fdhF,KSO-RS02125 and KSO-RS17170,which were involved in the transport of selenite,glucose metabolism,electron chain transfer,and secretion of SeNPs.When using 0.5 g∕L sodium selenite as the substrate,adding 20 g∕L glucose to LB medium had the best effect on promoting bacterial protein and SeNPs synthesis.At 72 hours,the conversion rate of SeNPs was 72.33%(control was 53.25%),and the expression levels of the above genes were significantly upregulated.In summary,the thioredoxin reduction system(TrxA,TrxB)and nitrite reductase(NfsA)might be more critical for the conversion of SeNPs in high concentration sodium selenite environments.Proper addition of glucose to LB medium could significantly increase the gene expression levels of key enzymes involved in SeNPs synthesis and the conversion efficiency of SeNPs.
