沉默lncRNA TUG1上调miR-139-5p表达增强肺癌A549细胞放射敏感性分析

摘　　要：目的探讨沉默lncRNA TUG1通过上调miR-139-5p表达增强肺癌A549细胞放射敏感性的效果。方法设A549肺癌细胞组、放疗(IR)组、lncRNA TUG1低表达(inhibitor)组、IR+lncRNA TUG1 inhibitor组。各组细胞培养结束后,采用MTT法测定细胞活力,甲基紫染色测定单克隆形成数目,流式细胞仪测定凋亡水平,Millipore Transwell小室测定侵袭水平,RT-PCR法及蛋白印迹法测定各组细胞lncRNA TUG1、miR-139-5p、SMC1A mRNA和蛋白水平。结果与A549肺癌细胞组比较,IR组、lncRNA TUG1 inhibitor组、IR+lncRNA TUG1 inhibitor组OD值、存活率、细胞克隆形成数目、穿膜数、lncRNA TUG1 mRNA表达水平、SMC1A mRNA和蛋白表达水平降低(P<0.05),凋亡率、miR-139-5p mRNA表达水平升高(P<0.05)。与IR组、lncRNA TUG1 inhibitor组比较,IR+lncRNA TUG1 inhibitor组OD值、存活率、细胞克隆形成数目、穿膜数、lncRNA TUG1 mRNA表达水平、SMC1A mRNA和蛋白表达水平降低(P<0.05),凋亡率、miR-139-5p mRNA表达水平升高(P<0.05)。结论沉默lncRNA TUG1能明显抑制肺癌A549细胞增殖、侵袭,促进A549细胞凋亡,增加A549细胞放射敏感性;这一效果可能与TUG1的敲低可以促进A549肺癌细胞高表达miR-139-5p进而抑制SMC1A的表达有关。Objective To investigate the effect of silencing lncRNA TUG1 on enhancing radiosensitivity of lung cancer A549 cells by up-regulating the expression of miR-139-5p.Methods A549 lung cancer cell group,radiotherapy(IR)group,lncRNA TUG1 low expression(inhibitor)group,IR+lncRNA TUG1 inhibitor group were set up.After the cell culture of each group was completed,the cell viability was determined by MTT method,the number of single clones formed by methyl violet staining,the level of apoptosis determined by flow cytometry,the level of invasion determined by Millipore Transwell chamber,and the level of lncRNA TUG1,miR-139-5p,SMC1A mRNA and protein levels in cells were determined by RT-PCR and Western blotting.Results Compared with the A549 lung cancer cell group,the OD value,survival rate,number of cell clones,number of penetrating membranes,lncRNA TUG1 mRNA expression level,SMC1A mRNA and protein expression levels in IR group,lncRNA TUG1 inhibitor group,and IR+lncRNA TUG1 inhibitor group decreased(P<0.05),apoptosis rate and miR-139-5p mRNA expression level increased(P<0.05).Compared with the IR group and the lncRNA TUG1 inhibitor group,the IR+lncRNA TUG1 inhibitor group had lower OD value,survival rate,number of cell clones,number of cell penetrating membrane,lncRNA TUG1 mRNA expression level,SMC1A mRNA and protein expression level(P<0.05),apoptosis rate and miR-139-5p mRNA expression level increased(P<0.05).Conclusion Silencing lncRNA TUG1 can significantly inhibit the proliferation and invasion of lung cancer A549 cells,promote the apoptosis of A549 cells,and increase the radiosensitivity of A549 cells.The mechanism may be related to that knockdown of TUG1 can promote the high expression of miR-139-5p in A549 lung cancer cells,thereby inhibiting SMC1A.
