摘 要:目的通过分析肾透明细胞癌组织中c-kit的表达情况,并对差异表达的c-kit进行相关生物信息学分析,进一步探讨烟草与c-kit在肾透明细胞癌患者中的潜在作用机制。方法利用癌症基因组图谱(the Cancer Genome Atlas,TCGA)数据库,分析c-kit在肿瘤组织与正常组织中的表达差异,Meta分析验证上述结果。基于UALCAN数据库,分析影响基因表达的临床病理特征。通过Linkedomics进行c-kit共表达基因的基因富集通路及PPI(protein-protein interaction)网络图的获取。Timer分析c-kit表达与肿瘤免疫浸润的关系。免疫组化分析肾透明细胞癌患者中c-kit蛋白的表达情况以及与吸烟史的相关性。结果c-kit在肾透明细胞癌中呈现低表达,且其表达与患者肿瘤分期相关。KIT在肾细胞癌的免疫浸润中发挥着重要的作用。肾透明细胞癌患者中c-kit阳性占10%,但其表达与吸烟无相关性。结论吸烟作为肾癌的重要危险因素,可能通过激活肿瘤组织中c-kit以外的其他信号通路在肾透明细胞癌的发生发展过程中发挥重要作用。Objective To explore the potential mechanism of smoking and c-kit in renal clear cell carcinoma patients by analyzing the expression of c-kit in clear cell renal cell carcinoma(ccRCC)tissues and the function of c-kit and associated genes through bioinformatics methods.Methods Utilizing the Cancer Genome Atlas(TCGA)database,we analyzed the differential expression of c-kit between tumor tissues and normal tissues,with meta-analysis conducted to validate these findings.Based on the UALCAN database,we explored the clinicopathological features influencing gene expression.Through LinkedOmics,we obtained gene enrichment pathways and Protein-Protein Interaction(PPI)network maps for c-kit co-expressed genes.TIMER analysis was employed to investigate the relationship between c-kit expression and tumor immune infiltration.Immunohistochemical analysis was performed to assess the expression of c-kit protein in patients with clear cell renal cell carcinoma and its correlation with smoking history.Results The expression of c-kit was significantly low in ccRCC,and its expression was correlated with tumor stage.The KIT was positive in 10%of patients,and the expression of KIT was not associated with smoking.Conclusion Smoking is an important risk factor of renal cell carcinoma and it may play an important role in the occurrence and development of ccRCC by activating other signaling pathways besides c-kit in tumor tissue.
