摘 要:目的研究钩藤37种加工方法的醇提物对DPPH自由基清除能力的影响。方法采用DPPH(1,1-二苯基-2-苦味基肼)自由基清除法,以抗坏血酸为阳性对照,采用紫外-可见分光光度仪测定吸光度,根据公式计算不同加工方法样品的IC_(50)值(清除率为50%时样品浓度),进一步评价钩藤不同加工方法的抗氧化活性。结果37种加工方法的醇提物清除自由基能力具有显著差异。作用较强的加工方法为蒸制法,IC_(50)值最低的依次为蒸制60 min,蒸1次(0.2438 mg/mL)<蒸制45 min,蒸1次(0.2591 mg/mL)<蒸制15 min,蒸1次(0.2613 mg/mL);而发汗法作用较弱,其中以直接发汗3天的抗氧化活性最差,IC_(50)值为0.4823 mg/mL。结论钩藤不同加工方法对DPPH自由基的清除能力不同,高温加热处理可提高其抗氧化活性,研究结果为该药材产地加工方法的综合质量评价及抗氧化活性研究提供重要参考。Objective To study the impact of alcohol-soluble extracts of 37 processing methods of Uncaria rhynchophylla on DPPH free radical scavenging capability.Methods The DPPH(1,1-diphenyl-2-picrylhydrazyl)free radical removal method was used,with Vitamin C serving as a positive control,and UV-visible spectrophotometry was employed to measure absorption.The IC_(50)value,defined as the sample concentration at which the removal rate is 50%,was calculated using a specific formula.This allowed for the further evaluation of the antioxidant activity of the different processing methods of Uncaria rhynchophylla.Results There is a significant difference in the free radical scavenging ability among the 37 processing methods.The steaming method demonstrated the strongest scavenging ability.The lowest IC_(50)value was found in the sample steamed for 60 minutes,once(0.2438 mg/mL),followed by the sample steamed for 45 minutes,once(0.2591 mg/mL),and the sample steamed for 15 minutes,once(0.2613 mg/mL).The sweating method showed weaker scavenging ability,with the antioxidant activity of 3 days of direct sweating being the poorest,having an IC_(50)value of 0.4823 mg/mL.Conclusion Different processing methods of Uncaria rhynchophylla exhibit varying abilities to scavenge DPPH free radicals.High-temperature heating treatments can enhance its antioxidant activity.These research results provide important references for the comprehensive quality evaluation and antioxidant activity of the habitat processing of Uncaria rhynchophylla.
