摘  要:目的:研究黄芩根型与其化学成分及抗氧化活性的相关性,探究不同根型黄芩抗氧化活性成分的差异。方法:以不同根型的黄芩为研究对象,建立其指纹图谱,测定有效成分含量,以1,1-二苯基-2-三硝基苯肼(DPPH)自由基清除法及2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS)自由基清除法的半抑制质量浓度(IC50)为抗氧化作用的药效指标,探究不同根型黄芩中发挥抗氧化活性的主要化学成分。采用灰色关联度分析、双变量相关性分析和偏最小二乘回归分析进行抗氧化活性的谱效关系研究,并对筛选的具有抗氧化活性的成分进行药效验证。结果:从不同根型黄芩的高效液相色谱指纹图谱中确定共有峰17个,与对照指纹图谱相似度不低于0.9000,并确认了黄芩苷(峰6)、汉黄芩苷(峰11)、黄芩素(峰14)、汉黄芩素(峰16)、白杨素(峰17)5个峰。直根小根型的黄芩有效成分含量较高。不同根型黄芩DPPH自由基清除法的IC50为0.112~0.168 mg/mL,ABTS自由基清除法的IC50为0.208~0.302 mg/mL。通过谱效关系分析筛选出峰2、峰3、峰6(黄芩苷)、峰7、峰10、峰11(汉黄芩苷)、峰14(黄芩素)可能为黄芩中具有抗氧化活性的成分。单体物质体外抗氧化活性成分验证结果显示,黄芩苷和黄芩素是清除DPPH自由基的主要物质,黄芩苷、黄芩素和汉黄芩素是清除ABTS自由基的主要物质。结论:不同根型黄芩有效成分含量有明显差异,直根小根型黄芩有效成分含量高,抗氧化能力强。通过单体化合物抗氧化活性验证得出黄芩苷和黄芩素是黄芩中具有抗氧化活性的主要物质,阐明了黄芩抗氧化的物质基础,为不同根型黄芩的质量控制与评价提供依据。Objective:To study the correlation between the root types of Huangqin(Scutellariae Radix)and its chemical components and antioxidant activity,and investigate the differences of antioxidant active components in different root types of Huangqin.Methods:Take different root types of Huangqin as the research object,the fingerprints were established to determine the content of effective components.The main chemical components of different root types of Huangqin were studied using the semi-inhibitory concentration(IC50)of free radical scavenging of 1,1-diphenyl-2-picrylhydrazyl radica(l DPPH)and 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid ammonium salt)(ABTS)as the pharmacodynamic index of antioxidant activity.Grey correlation analysis,bivariate correlation analysis and partial least squares regression analysis were used to study the spectrum-effect relationship of antioxidant activity,and the efficacy of the selected antioxidant components was verified.Results:A total of 17 peaks were identified from the HPLC fingerprints of different root types of Huangqin,with a similarity of no less than 0.9000 to the control fingerprint.Five peaks of baicalin(peak 6),wogonoside(peak 11),baicalein(peak 14),wogonin(peak 16),and chrysin(peak 17)were also identified.The content of effective components in Huangqin with taproot and rootlet was relatively high.The IC50 of free radical scavenging of DPPH and ABTS in different root types of Huangqin were 0.112~0.168 mg/mL and 0.208~0.302 mg/mL,respectively.Through the analysis of spectrum-effect relationship,peak 2,peak 3,peak 6(baicalin),peak 7,peak 10,peak 11(wogonin)and peak 14(baicalein)in the chromatogram were identified as potential active components with antioxidant activity in scutellaria baicalensis.The results of validation of antioxidant active components in vitro showed that baicalin and baicalein were the main substances for free radical scavenging of DPPH.Baicalin,baicalein and wogonin were the main substances for free radical scavenging of ABTS.Conclusions:Differen
