摘　　要：目的分析miR-28-3p在肝细胞癌组织中的表达与患者临床病理特征及预后的关系,研究其对肝细胞癌细胞增殖、侵袭和迁移的影响。方法从TCGA数据库下载肝细胞癌miR-28-3p表达数据、患者临床病理参数及生存预后信息,分析miR-28-3p与肝细胞癌患者临床病理参数及预后之间的关系;使用化学合成的miR-28-3p mimics及mimics NC转染肝细胞癌细胞株Huh7,用CCK-8和Transwell侵袭和迁移实验检测各组细胞的增殖、侵袭和迁移能力。结果MiR-28-3p在具有肝纤维化背景的肝细胞癌组织中高表达,其表达水平与肿瘤大小、TNM分期显著相关(P<0.05),miR-28-3p高表达者预后更差(P<0.05)。体外细胞实验表明,与mimics NC组比较,miR-28-3p mimics转染组肝细胞癌细胞增殖、侵袭和迁移能力均显著提高(均P<0.05)。结论MiR-28-3p与肝细胞癌的分期显著相关,是患者不良预后的独立预测指标,其高表达促进肝细胞癌细胞的增殖、侵袭和迁移能力。Objective To study the relationship between miR-28-3p expression in hepatocellular carcinoma(HCC)and HCC patients'clinicopathological characteristics and prognosis,and then to explore the effect of miR-28-3p expression on the proliferation,invasion,and migration of HCC cells.Methods MiR-28-3p expression data of HCC,clinicopathological parameters,and survival information were downloaded from the Cancer Genome Atlas(TCGA)database,and the relationships between miR-28-3p expression and clinicopathological parameters and prognosis were analyzed respectively.Chemically synthesized miR-28-3p mimics and negative control were used to transfect HCC cells line Huh7,and then cell counting Kit-8(CCK-8)and tranwell invasion and migration assays were employed to detect the proliferation,invasion and migration abilities of the cells in each group.Results MiR-28-3p was highly expressed in HCC tissues with a liver fibrosis background,and its expression level was significantly correlated with tumor size and TNM stage(P<0.05).Patients with high expression of miR-28-3p had a worse prognosis(P<0.05).As Cox analysis shows,miR-28-3p is an independent prognostic factor for patients with HCC.Vitro cell experiments found that,compared with the mimics NC,transfection of miR-28-3p mimics can significantly improved the proliferation,invasion,and migration abilities of HCC cells(all P<0.05).Conclusion MiR-28-3p is an independent predictor of poor prognosis of HCC,and high expression of miR-28-3p promotes the proliferation,invasion,and migration capabilities of HCC cells.
