摘  要:目的探讨土贝母苷甲(TBMS I)对肝癌细胞增殖的影响及其分子机制。方法采用MTS实验和克隆形成实验检测不同浓度TBMS I对肝癌细胞株MHCC97-H生长的影响。采用检测葡萄糖摄取、乳酸生成、乳酸脱氢酶(LDH)活性、细胞外液pH、细胞氧耗和线粒体呼吸链复合体Ⅳ活性的实验,分析TBMS I对肝癌细胞糖酵解、氧化磷酸化的影响。采用qRT-PCR和Western blot实验检测肝癌细胞株MHCC97-H中缺氧诱导因子-1α(HIF-1α)的表达。结果TBMS I作用使肝癌细胞的增殖能力下降,且呈一定的剂量和时间依赖效应;使葡萄糖摄取和乳酸生成减少,LDH活性降低,细胞外培养液pH升高,细胞氧耗和线粒体呼吸链复合体Ⅳ活性增加。结论TBMS I通过抑制HIF-1α的表达抑制细胞糖酵解和促进氧化磷酸化,进而抑制肝癌细胞增殖。Objective This study aims to investigate the impact of Tubeimoside I on the proliferation of liver cancer cells and its molecular mechanism.Methods MTS assay and colony formation assay were conducted to examine the effects of different concentrations of Tubeimoside I on the growth of MHCC97-H liver cancer cells.Glucose uptake,lactate production,LDH activity,extracellular pH,cellular oxygen consumption and mitochondrial respiratory chain complex IV activity were measured to analyze the influence of Tubeimoside I on glycolysis and oxidative phosphorylation in liver cancer cells.qRT-PCR and Western blot were performed to detect the expression of HIF1αin MHCC97-H liver cancer cells.Results Tubeimoside I treatment decreased the proliferation ability of liver cancer cells in a dose-and time-dependent manner.It also reduced glucose uptake,lactate production and LDH activity,increased extracellular pH,and enhanced cellular oxygen consumption and mitochondrial respiratory chain complex IV activity.Conclusion Tubeimoside I inhibits cell glycolysis and promotes oxidative phosphorylation by suppressing the expression of HIF-1α,thereby suppressing the proliferation of liver cancer cells.
