不同类型返蓝液在苏木精-伊红切片中的应用及对切片褪色的影响研究

摘　　要：目的探讨不同类型返蓝液在苏木精-伊红(HE)切片中的应用对比及对切片褪色的分析。方法选择2021年10月至2022年3月期间南平市第二医院病理科选取的30个阑尾蜡块,20个乙状结肠癌蜡块、23个扁桃体蜡块,20个胃黏膜活检蜡块,总共93个蜡块。93个蜡块均行制备HE切片,每个蜡块连续切片均切3张同样厚度切片,分别分到3组中,每组都含以上93个蜡块中的1张同等厚度的切片。对照1组采用饱和碳酸锂返蓝,对照2组采用稀氨水返蓝,观察组采用EDTA返蓝,比较3组HE染色情况、HE切片褪色情况。结果3组在组织结构、细胞核形态、细胞质染色情况、细胞核染色及核质对比评分存在差异(P<0.05)。观察组组织结构、细胞核形态、细胞质染色情况、细胞核染色及核质对比评分分别高于对照1组和对照2组(P<0.05)。3组在染色后的第30天、第90天、第180天、第270天、第360天、第390天切片褪色情况评分存在差异(P<0.05)。观察组在染色后的第30天、第90天、第180天、第270天、第360天、第390天切片褪色情况评分分别高于对照1组和对照2组(P<0.05)。结论不同类型返蓝液用于HE切片中均可获得良好的染色情况,但是EDTA返蓝染色质量好对切片褪色影响较小,方便后期复片。Objective To explore the application comparison of different types of bluing solutions in Hematoxylin-Eosin(HE)sections and to analyze the fading of sections.Methods From October 2021 to March 2022,30 appendiceal paraffin blocks,20 sigmoid colon cancer paraffin blocks,23 tonsil paraffin blocks,and 20 gastric mucosal biopsy paraffin blocks were selected from the Department of Pathology at Nanping Second Hospital,totaling 93 paraffin blocks.HE sections were prepared from all 93 paraffin blocks,and each block was cut into 3 sections of the same thickness,which were then divided into 3 groups,each containing one section of the same thickness from the 93 blocks.Group 1 used saturated lithium carbonate for bluing,Group 2 used dilute ammonia water for bluing,and the observation group used EDTA for bluing.The HE staining situation and the fading of HE sections were compared among the three groups.Results There were differences in tissue structure,cell nucleus morphology,cytoplasm staining,nuclear staining,and nuclear-cytoplasmic contrast scoring among the three groups(P<0.05).The observation group had higher scores in tissue structure,cell nucleus morphology,cytoplasm staining,nuclear staining,and nuclear-cytoplasmic contrast than Groups 1 and 2(P<0.05).There were differences in the fading scores of the sections on the 30th,90th,180th,270th,360th,and 390th days after staining among the three groups(P<0.05).The observation group had higher fading scores on the 30th,90th,180th,270th,360th,and 390th days after staining than Groups 1 and 2(P<0.05).Conclusions Different types of bluing solutions can achieve good staining results in HE sections,but EDTA bluing has better staining quality and less impact on section fading,which is convenient for later review of slides.
