摘  要:毛木耳是我国重要的商业食用菌之一,具有较高的食药用价值。为了探究毛木耳不同出菇时期的转录本差异表达情况,本文以漳耳43-28为研究材料,通过高通量测序获得不同出菇时期的差异表达基因,并对其进行GO功能富集和KEGG富集分析。GO功能富集发现,差异表达基因主要富集在代谢进程、细胞进程和膜组成等与代谢和细胞组成相关的进程。KEGG富集发现,原基与小耳时期的差异表达基因显著性富集在DNA修复、同源重组、细胞周期-酵母菌、淀粉和蔗糖代谢、糖酵解/糖原生成、色氨酸代谢、酪氨酸代谢和苯丙氨酸代谢等与细胞增殖、能量代谢和黑色素合成相关的途径;小耳与成熟时期的差异表达基因显著性富集在酪氨酸代谢和与能量代谢相关的氧化磷酸化。由此可推测,与细胞增殖、能量代谢和黑色素合成相关的代谢通路为毛木耳的原基分化和子实体成熟提供能量和物质基础。Auricularia cornea is one of the important commercial edible fungi in China,with high medicinal and food value.In order to investigate the differential expression of transcripts in different fruiting stages of A.cornea,the cultivated strain‘Zhang’er 43-28’was used as research material,and the differentially expressed genes in different fruiting stages were obtained by high-throughput sequencing.The analyses of GO function enrichment and KEGG enrichment of the genes were carried out.GO function enrichment found that the differentially expressed genes were mainly enriched in the metabolic process,cellular process,and membrane part,which were related to metabolism and cellular composition.KEGG enrichment revealed that the differentially expressed genes of the primordium formation and initial fruiting stages were significantly enriched in DNA replication,homologous recombination,cell cycle-yeast,starch and sucrose metabolism,glycolysis/glycogenesis,tryptophan metabolism,tyrosine metabolism,phenylalanine metabolism,and the others related to the pathways of cell proliferation,energy metabolism and melanin synthesis.The differentially expressed genes in the initial fruiting and fruiting body maturation stages were mainly enriched in oxidative phosphorylation related to energy metabolism and tyrosine metabolism.It can be inferred that the metabolic pathways related to cell proliferation,energy metabolism and melanin synthesis provide the energy and material basis for the primordial polarization and fruiting body maturation of A.cornea.
