摘 要:目的:探讨抗缪勒管激素(AMH)对卵巢癌OVCAR8细胞迁移和侵袭的影响及其作用机制。方法:采用0、5、15、25nM AMH处理人卵巢癌OVCAR8细胞,相应分为对照组、AMH 5nM组、AMH 15nM组和AMH 25nM组。通过划痕实验和Transwell实验分别检测各组OVCAR8细胞迁移和侵袭能力,运用Western Blotting检测各组OVCAR8细胞Wnt/β-catenin途径相关分子蛋白水平,采用免疫荧光实验检测各组OVCAR8细胞肿β-catenin荧光表达情况。采用Wnt/β-catenin通路激活剂SKL2001和AMH(抑制效应最高的浓度25nM)共同作用OVCAR8细胞(AMH 25nM+SKL2001组)后检测细胞迁移和侵袭的变化。结果:AMH能够以浓度依赖的方式抑制OVCAR8细胞的迁移和侵袭。AMH呈浓度依赖性降低OVCAR8细胞Wnt1、β-catenin、C-Myc、Cyclin D1蛋白表达水平及细胞β-catenin荧光表达水平。采用Wnt/β-catenin通路激活剂SKL2001作用细胞后,OVCAR8细胞迁移和侵袭细胞数及Wnt1、β-catenin、C-Myc、Cyclin D1蛋白表达水平显著升高(P<0.01)。结论:AMH能够抑制卵巢癌细胞迁移和侵袭,其作用机制可能与抑制Wnt/β-catenin信号通路有关。Objective:To investigate the effects of human anti-Müllerian hormone(AMH)on the migration and invasion of ovarian cancer OVCAR8 cells and its mechanism.Method:OVCAR8 cells were treated with 0,5,15,25nM AMH and divided into control group,AMH 5nM group,AMH 15nM group and AMH 25nM group.The migration and invasion ability of OVCAR8 cells in each group were detected by scratch test and Transwell test,and the Wnt/β-catenin pathway molecular protein levels of OVCAR8 cells in each group were detected by Western Blotting.The expression ofβ-catenin in OVCAR8 cells was detected by immunofluorescence assay.Wnt/β-catenin pathway activator SKL2001+AMH were used to detect the changes of cell migration and invasion.Results:AMH inhibited OVCAR8 cell migration and invasion in a concentration-dependent manner.AMH reduced the expression levels of Wnt1,β-catenin,C-Myc,Cyclin D1 andβ-catenin fluorescence in OVCAR8 cells in a concentration-dependent manner.After the treatment of Wnt/β-catenin pathway activator SKL2001,OVCAR8 cell migration and invasion,and the protein expression levels of Wnt1,β-catenin,C-Myc,Cyclin D1 were significantly increased(P<0.01).Conclusion:AMH can inhibit the migration and invasion of ovarian cancer cells,and its mechanism may be related to the inhibition of the Wnt/β-catenin signaling pathway.
