胶质瘤患者LncRNA SNHG25、miR-497-5p水平及其与临床病理特征和预后的相关性研究

摘　　要：目的:探讨胶质瘤组织LncRNA SNHG25、miR-497-5p表达与患者临床病理特征及预后的相关性。方法:选取本院2018-01-2020-03收治的84例胶质瘤患者为观察组,并同期选取84例因颅脑损伤并接受手术治疗的患者为对照组。分别手术切除胶质瘤组织和正常脑组织后,比较两组LncRNA SNHG25、miR-497-5p水平。比较观察组中不同临床特征患者LncRNA SNHG25、miR-497-5p水平的差异。采用Pearson相关法分析两组患者LncRNA SNHG25水平与miR-497-5p水平相关性。对观察组患者随访3年,根据是否死亡分为死亡组和存活组,比较两组LncRNA SNHG25、miR-497-5p水平,通过受试者工作特征(ROC)曲线分析LncRNA SNHG25、miR-497-5p水平对胶质瘤患者预后预测的临床价值。结果:与对照组比较,观察组LncRNA SNHG25水平升高,miR-497-5p水平降低(P<0.05)。在胶质瘤患者中,与肿瘤直径<4cm、WHO分级为Ⅰ-Ⅱ级的患者比较,肿瘤直径≥4cm、WHO分级为Ⅲ-Ⅳ级患者LncRNA SNHG25水平升高,miR-497-5p水平降低(P<0.05);相关性分析结果显示,胶质瘤患者LncRNA SNHG25水平与miR-497-5p水平呈负相关(r=-0.369,P<0.01),ROC曲线结果显示,当LncRNA SNHG25截断值为1.83时,预测胶质瘤患者预后的AUC为0.86;当miR-497-5p截断值为0.91时,预测胶质瘤患者预后的AUC为0.91。结论:胶质瘤患者LncRNA SNHG25和miR-497-5p水平与肿瘤直径、分期有关,二者对辅助评估预后具有一定临床价值。Objective:To investigate the correlation between the expressions of LncRNA SNHG25 and miR-497-5p in glioma tissue and clinicopathological features and prognosis.Method:A total of 84 glioma patients were selected as research objects,and 84 patients who received surgical treatment in our hospital due to craniocerebral injury were selected as control group.After resection of glioma tissue and normal brain tissue,LncRNA SNHG25 and miR-497-5p levels were compared between the two groups.The observation group was divided into different subgroups according to clinical characteristics,and the levels of LncRNA SNHG25 and miR-497-5p among subgroups were compared.Correlation method was used to analyze the correlation between LncRNA SNHG25 level and miR-497-5p level in the two groups.Patients in the observation group were followed up for 3 years and divided into death group and survival group according to whether they died or not.The levels of LncRNA SNHG25 and miR-497-5p in the two groups were compared,and the clinical value of LncRNA SNHG25 and miR-497-5p in predicting prognosis was analyzed by ROC curve.Results:Compared with control group,LncRNA SNHG25 level in observation group was increased,and miR-497-5p level was decreased(P<0.05).Compared with glioma patients with tumor diameter<4cm and WHO grade I-II,LncRNA SNHG25 levels were increased and miR-497-5p levels were decreased in patients with tumor diameter≥4cm and WHO grade III-IV(P<0.05).Correlation analysis results showed that LncRNA SNHG25 levels were negatively correlated with miR-497-5p levels in glioma patients(r=-0.369,P<0.01).ROC curve results showed that when the cut-off value of LncRNA SNHG25 was 1.83,the AUC for predicting the prognosis of glioma patients was 0.86,and when the cut-off value of miR-497-5p was 0.91,the AUC for predicting the prognosis of glioma patients was 0.91.Conclusion:The levels of LncRNA SNHG25 and miR-497-5p were related to tumor diameter and stage,which had certain clinical value in assisting prognosis assessment.
