摘 要:目的探究莱菔子提取物在糖尿病牙周炎大鼠模型中对牙周炎症的改善作用并分析其与PI3K/AKT信号通路之间的关联性。方法40只SD雄性大鼠随机等量分为5组:空白对照组(N组)、牙周炎模型组(P组)、牙周炎莱菔子治疗组(P+Q组)、糖尿病牙周炎模型组(DP组)、糖尿病牙周炎莱菔子治疗组(DP+Q组)。除N组外,其余4组大鼠均通过口腔正畸丝牙周结扎法与菌液涂布法联合建立牙周炎模型;DP组和DP+Q组辅以一次性腹腔注射链脲佐菌素建立糖尿病牙周炎模型。建模成功后,P+Q组和DP+Q组通过灌胃给药的方式分别给予莱菔子提取物2.5g/(kg·d),N组、P组和DP组则给予等量生理盐水。连续给药8周,过程中观察大鼠一般状况并跟踪大鼠的空腹血糖变化。给药结束后处死取材,处死前记录大鼠牙龈指数、龈沟出血指数;记录牙槽骨吸收高度;利用酶联免疫吸附法检测血清中白细胞介素-6和肿瘤坏死因子-α水平;采用苏木精-伊红染色观察并比较大鼠牙周组织病理形态变化;运用Western blot法测定大鼠牙周组织中PI3K/AKT信号通路中相关蛋白的表达水平。结果P组和DP组白细胞介素-6和肿瘤坏死因子-α水平显著高于N组,DP组白细胞介素-6和肿瘤坏死因子-α水平显著高于P组(P<0.05)。P+Q组和DP+Q组白细胞介素-6和肿瘤坏死因子-α水平分别低于P组和DP组(P<0.05)。DP组PI3K、AKT表达量显著低于N组(P<0.05)。DP+Q组PI3K、AKT表达量显著高于DP组(P<0.05)。结论莱菔子提取物可基于PI3K/AKT信号通路改善糖尿病牙周炎大鼠炎症反应,保护牙周组织。Objective To explore the protective effect of Raphani semen extract on periodontal inflammation in diabetic periodontitis rat model and analyze the correlation between Raphani semen extract and PI3K/AKT signaling pathway.Methods Forty male Sprague-Dawley(SD)rats were selected using random sampling methods and then the subjects were initially divided into five groups and then categorized as follows:the blank control group(Group N),periodontitis control group(Group P),periodontitis Raphani semen treatment group(Group P+Q),diabetic periodontitis group(Group DP),and diabetic periodontitis Raphani semen treatment group(Group DP+Q),with eight animals in each group.Except group N,the periodontitis model of the other 4 groups was established by orthodontic wire periodontitis ligation combined with bacterial solution coating.Specifically,the DP group and DP+Q group received an additional intraperitoneal one-time injection of streptozotocin to establish the diabetic periodontitis rat model.After successful modeling,Raphani semen extract 2.5g/(kg·d)was administered by gavage in the P+Q and DP+Q groups,respectively,and an equal amount of saline was given to the N,P,and DP groups.The drug was administered continuously for 8 weeks,during which the general state of the rats was observed and the changes of fasting blood glucose in the rats were tracked.Gingival index(GI)and sulcus bleeding index(SBI)of rats were recorded before execution.After execution,the height of alveolar bone resorption was recorded;serum IL-6 and TNF-αlevels were assessed using ELISA,while pathological and morphological alterations in rat periodontal tissues were examined via HE staining.and protein expression levels of PI3K and AKT in rat periodontal tissues were detected by Western blotting.Results The inflammation levels in the P and DP groups were significantly higher than those in the N group(P<0.05),with those in the DP group significantly higher than those in the P group(P<0.05).The inflammation level of the P+Q and DP+Q groups treated with Raphan
