摘 要:目的 制备一种能实现顺序释放药物的芯壳静电纺丝短纤维及其混悬液用于局部注射使用,并观察该短纤维对人类非小细胞肺癌细胞系A549及其裸鼠移植肿瘤的作用。方法 观察不同(SF-HD:壳层羟喜树碱,芯层阿霉素;SF-DH:壳层阿霉素,芯层羟喜树碱)短纤维的结构及药物释放特点,用各组短纤维分别处理A549细胞,采用实时细胞分析技术及流式技术观察其生长和凋亡状况。裸鼠右后肢上方接种A549细胞,造模成功后随机分为空白组、未载药短纤维对照组、SF-HD、SF-DH组,于肿瘤中心上方分别注射200μl的0.9%氯化钠溶液,未载药短纤维混悬液、SF-DH混悬液、SF-HD混悬液,注射日为第0天,每隔两天测量肿瘤体积和小鼠体质量,并绘制成曲线。注射5、10、21天后,取肿瘤组织,苏木精-伊红(hematoxylin eosin, HE)染色观察。结果 与SF-DH比较,SF-HD在抑制A549细胞及其移植肿瘤的生长、促进其凋亡的作用更明显。结论SF-HD组对A549细胞及其移植瘤具有更好的抑制效果。本研究中制备的静电纺丝短纤维缓释系统可随时间推移实现缓慢控释的效果,是具有研究前景的给药系统。Objective To prepare a core-shell electrostatically spun short fiber capable of sequential drug release and its suspension for local injection use,and to observe the effects of the short fiber on human non-small cell lung cancer cell line A549 and its transplanted tumor in nude mice.Methods The structure and drug release characteristics of different(SF-HD:shell hydroxycamptothecin,core adriamycin;SF-DH:shell adriamycin,core hydroxycamptothecin)staple fibers were observed.A549 cells were treated with each group of staple fibers,and real-time cellular analysis and flow technology were used to observe growth and apoptosis.Nude mice were inoculated with A549 cells above the right hind limb,and after successful modeling,they were randomly divided into blank,control,SF-HD,and SF-DH groups,and injected with 200ul of saline,blank short fiber suspension,SF-DH suspension,and SF-HD suspension above the center of the tumor,respectively.The day of injection was 0 day,and the volume of the tumors and the weight of the mice were measured every two days and plotted as a curve.After 5,10 and 21 days of injection,tumor tissues were taken and observed by HE staining.Results Compared with SF-DH,SF-HD had more obvious effects in inhibiting the growth and promoting the apoptosis of A549 cells and their transplanted tumors.Conclusion The SF-HD group had a better inhibitory effect on A549 cells and their transplanted tumors.The slow-release system of electrostatically spun short fibers prepared in this study can achieve the effect of slow controlled release over time and is a promising drug delivery system for research.
